Antibody Response of Adults to an Aluminum Hydroxide-Adsorbed Neisseria meningitidis Serotype 2b Protein-Group B Polysaccharide Vaccine

Abstract: A group B Neisseria meningitidis serotype protein vaccine was studied clinically in adults. The vaccine comprised lipopolysaccharide-depleted outer membrane vesicles from a serotype 2b strain, 3006-M2, noncovalently complexed with group B meningococcal polysaccharide. Volunteers received 25 micrograms each of protein and polysaccharide administered intramuscularly either in 0.9% NaCl or adsorbed onto aluminum hydroxide on weeks 0 and 6. Most individuals experienced mild local reactions, but there were no syste… Show more

“…1C and 1D. These results indicated that the blue color resulting from lactose was similar to that resulting from BSA, and thus confirmed that lactose interferes with the assay, as previously described by Frasch et al (14). In addition, these results revealed that lactose in samples should be removed in order to accurately determine the protein content.…”

“…Lactose has been previously reported to interfere with the Lowry assay of a vaccine (14), but the degree to which lactose interfered with the Lowry assay was unclear. In this study, it was demonstrated that AE0.1 mg/mL lactose caused interference.…”

“…Da mesma forma, o polissacáride B purificado e de alto peso molecular induz primariamente à produção de IgM 19,31 , mesmo quando conjugado a proteínas carreadoras e adsorvidos ao hidróxido de alumínio 19,35 . Esta pobre imunogenicidade é atribuída, principalmente, à semelhança antigênica do polissacáride B com glicolípides e glicoproteínas presentes em células humanas, à sensibilidade a neuraminidases do organismo humano e a instabilidade tridimensional da molécula 31 .…”
