Background Paromomycin is a 2-deoxystreptamine aminocyclitol aminoglycoside antibiotic with broad spectrum activity against Gram-negative, Gram-positive bacteria and many protozoa. This study introduces a strategy for paromomycin production through solid-state fermentation using Streptomyces rimosus subsp. paromomycinus NRRL 2455. Solid state fermentation has gained enormous attention in the development of several products because of their numerous advantages over submerged liquid fermentation. After selecting the best solid substrate, a time course study of paromomycin production was carried out followed by optimization of environmental conditions using response surface methodology. Paromomycin yields obtained using this technique were also compared to those obtained using submerged liquid fermentation. Results Upon screening of 6 different substrates, maximum paromomycin concentration (0.51 mg/g initial dry solids) was obtained with the cost-effective agro-industrial byproduct, corn bran, impregnated with aminoglycoside production media. Optimization of environmental conditions using D-optimal design yielded a 4.3-fold enhancement in paromomycin concentration reaching 2.21 mg/g initial dry solids at a pH of 8.5, inoculum size of 5% v/w and a temperature of 30 °C. Conclusion Compared to submerged liquid fermentation, solid state fermentation resulted in comparable paromomycin concentrations, cost reduction of raw materials, less energy consumption and waste water discharge, which have major implications in industrial fermentation. Therefore, solid state fermentation is a promising alternative to submerged liquid fermentation for paromomycin production. To the best of our knowledge, this is the first report on the optimized paromomycin production through solid state fermentation process.

Streptomyces has been reported as an essential producer of bioactive substances, including antibiotics and other types of antimicrobials. This study investigated antibacterial-producing Streptomyces isolated from the gut of estuarine fish Chanos chanos, emphasizing screening for the producer of peptide-containing antibacterial compounds. Eighteen isolates were found during preliminary screening, in which four isolates showed the best antibacterial activities. Based on the morphological, physiological, and biochemical characterization, as well as 16S rRNA partial sequencing, all of the four isolates belonged to Streptomyces. Three isolates were suspected as novel isolate candidates based on homology presentations and phylogenetic tree analysis. Disk-diffusion assay of the metabolite-crude-extract from the isolates showed broad-spectrum inhibitory activities against Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 10876, Escherichia coli ATCC 25922, and Pseudomonas aeruginosa InaCC B52 with minimum inhibitory concentration and minimum bactericidal concentration ranging from 2.5–10 mg/mL and 5–10 mg/mL, respectively. The highest antibacterial activity with low MIC and MBC values was shown by isolate AIA-10. Qualitative HPLC profiling revealed that the metabolic-crude-extracts showed many peaks with intensive area at 210 and 214 nm, especially from SCA-11 and AIA-10, indicating the presence of peptide groups in the structure of the constituent compound. The results also suggested that crude extracts SCA-11 and AIA-10 had higher hydrophobicity properties than the other extracts. Further characterization of the active compound was needed to find out which compounds were responsible for the antibacterial activity. The results of this study indicated that some Streptomyces isolated from new environmental niches, i.e., gut of estuarine fish Chanos chanos, produce promising peptide-containing bioactive compounds.

Abstract. Rozirwan, Muda HI, Ulqodry TZ. 2020. Short Communication: Antibacterial potential of Actinomycetes isolated from mangrove sediment in Tanjung Api-Api, South Sumatra, Indonesia. Biodiversitas 21: 5723-5728. Actinomycetes play an important role in the decomposition process of organic matter in mangrove sediments. This study was carried out to identify and evaluate the antagonistic activity of actinomycetes from mangrove sediment against Escherichia coli and Staphylococcus aureus. Identification of actinomycetes was performed based on morphological dan physiological characters. The antagonistic test was carried out against Escherichia coli and Staphylococcus aureus. A total of ten isolates had been successfully isolated and grouped into eight genera, including; Oerskovia H1, Micrococcus H2, Nocardia H3, Sporichtya H5, Corynebacterium H6, Jonesia H7, Actinomyces H10, and Streptomyces H8, H11, H12. Actinomycetes from mangrove sediment were gram-positive bacillus. The shape of the colony varied from circular to filamentous and irregular, with medium and large colony sizes. The colony appears white or yellow. The surface is convex and flat, aerobes and facultative anaerobes; capable of fermenting glucose. Five isolates have very strong growth inhibition activity against E. coli: Nocardia H3 (27.81mm ± 1.39), Actinomycetes H10 (27.75mm ± 2.48), Corynebacterium H6 (27.69mm ± 2.39), Micrococcus H2 (18.55mm ± 2.44), and Streptomyces H8 (17.92mm ± 2.70) while Streptomyces H8 (24.83mm ± 2.08), and Actinomycetes H10 (16.06mm ± 2.49) were active against S. aureus.
