Plants of the genus Hypericum, long used in folk medicine, contain active compounds which present, anti-septic, diuretic, digestive, expectorant, vermifugal, anti-depressive and other properties. The possible clastogenic effect of a H. brasiliense extract was tested in vivo on the bone marrow cells of Wistar rats. The extract was administered by gavage at doses of 50, 150 and 300 mg/kg body weight. Experimental and control animals were submitted to euthanasia 24 h after the treatment for micronucleus (MN) and chromosome preparations. H. brasiliense extract did not induce statistically significant increases in the average numbers of MN or chromosome aberrations in the test systems employed.

Three phloroglucinols were obtained from Hypericum brasiliense: japonicine A (1), isouliginosin B (2) and uliginosin B (3). Bioautography and disk diffusion methods were used to determine antibacterial activity of the hexanic extract. Strains of the Coagulase Negative Staphylococcus and American Methicillin Resistant Staphylococcus aureus clones showed a growth inhibition zone ranging from 10 to 12 mm and 7 to 15 mm, respectively. Minimal inhibitory concentration (MIC) values were used to measure antistaphylococcal activity for all phloroglucinols. Isouliginosin B and uliginosin B presented MIC values of 1.5 and 3.0 µg/mL, respectively, while japonicine A displayed MIC value of 50.0 µg/mL. Keywords: Hypericum brasiliense; antibacterial activity; phloroglucinol. O gênero Hypericum é representado por 420 espécies pertencentes à família das Clusiaceae (Guttiferae). Esta família é composta por espécies com importantes efeitos farmacológicos e biológicos tais como antibacteriano, antiviral, antioxidante, atividade citótoxica e antidepressiva. 3,4 O Hypericum perforatum é a espécie medicinal mais reconhecida, amplamente estudada e utilizada no tratamento de depressão leve a moderada. 5 Alguns estudos clínicos e experimentais propõem que o mecanismo de ação parece estar relacionado com o bloqueio não seletivo da recaptação de serotonina, noradrenalina e dopamina. Os efeitos antidepressivos são devidos especialmente à hiperforina, um derivado de floroglucinol. 6 Outros derivados de floroglucinóis, conjugados com o ácido filicínico, demonstraram excelente atividade antiestafilocócica, podendo-se considerar o gênero Hypericum como uma fonte de moléculas com potencial atividade antibacteriana. 7 No Brasil, a família Clusiaceae é representada por 21 gêneros e 183 espécies, sendo o Hypericum brasiliense o mais estudado. 4 Esta planta é um arbusto bianual, encontrada nas regiões sul e sudeste do Brasil 8 e é usada na medicina tradicional com propriedades antiespasmódicas e no tratamento de doenças infecciosas. 9 O H. brasiliense é quimicamente constituído de flavonóides, xantonas e floroglucinóis derivados do ácido filicínico com significativos efeitos farmacológicos e biológicos. Neste trabalho, verificou-se a aplicabilidade de uma metodologia computacional para se predizer a estrutura de compostos orgânicos com atividade biológica. Para isso, selecionaramse três floroglucinóis, e compararam-se suas conformações obtidas por modelagem molecular e por difração de raios X. Os resultados mostraram que as conformações obtidas por análise conformacional com o método AM1 seguidas de otimização de geometria utilizando o método DFT (B3LYP/6-31G(d,p)) estão em boa concordância com os dados obtidos experimentalmente por difração de raios X, indicando que a metodologia empregada parece ser uma ótima ferramenta para predizer preferências conformacionais desta classe de compostos.In this work we intend to verify the applicability of a computational methodology to predict structural features of organic compounds with biological activity. We selected three phloroglucinols and compared their calculated conformational data with their X-ray crystallographic structure. The results showed that conformations obtained by conformational analysis with the AM1 method followed by geometry optimization by using the DFT B3LYP/6-31 G(d,p) basis set are in very good agreement with X-ray data, indicating that the methodology employed here seems to be a very useful tool in order to predict the conformational preference for this class of compounds.Keywords: conformational analysis, X-ray, molecular modeling, phloroglucinols IntroductionDespite the development of organic synthesis, biotechnology and combinatorial chemistry, natural plants are still a great source of bioactive compounds.
