The antibacterial mechanism of action of Caesalpinia bonducella seed oil on membrane permeability of Listeria monocytogenes NCIM 24563 (MIC: 2 mg/mL) and Escherichia coli ATCC 25922 (MIC: 4 mg/mL) was determined by measuring the extracellular ATP concentration, release of 260-nm absorbing materials, leakage of potassium ions and measurement of relative electrical conductivity of the bacterial cells treated at MIC concentration. Its mode of action on membrane integrity was confirmed by release of extracellular ATP (1.42 and 1.33 pg/mL), loss of 260-nm absorbing materials (4.36 and 4.19 optical density), leakage of potassium ions (950 and 1000 mmol/L) and increase in relative electrical conductivity (12.6 and 10.5%) against food-borne pathogenic bacteria L. monocytogenes and E. coli, respectively. These findings propose that C. bonducella oil compromised its mode of action on membrane integrity, suggesting its enormous food and pharmacological potential.

Background: A great revival of scientific interests in drug discovery has been witnessed in recent years from medicinal plants for health maintenance. The aim of this work was to investigate three Nigerian medicinal plants collected in Nigeria for their in vitro antiplasmodial and antimicrobial activities. Materials and Methods: Extracts obtained from parts of Persea americana, Jatropha podagrica and Picralima nitida and their fractions were evaluated for in vitro antiprotozoal and antimicrobial activity. Result: The methanol extract of P. nitida demonstrated activity against chloroquine-sensitive and chloroquine-resistant P. falciparum clones with IC 50 values of 6.3 and 6.0 µg/mL, respectively. Methanol and chloroform extracts of P. americana seed showed antifungal activity against Cryptococcus neoformans IC 50 less than 8 and 8.211 µg/mL respectively. Finally, the petroleum ether extract of P. americana had activity against methicillinresistant Staphylococcus aureus (MRSA) with an IC 50 value of 8.7 µg/mL. Conclusion: The study revealed the antibacterial and antiplasmodial activities of the plants extracts at the tested concentrations.

Jatropha multifida LINN (Euphorbiaceae), a shrub, is found in different parts of India. The plant is known to exhibit antibiotic activity.2) Earlier examination of the latex of the plant afforded some cyclic peptides, phenolics and glucosides. [3][4][5] However, the investigation on other parts of the plant has not yet been reported.In continuation of our work 6-9) on the constituents of Jatropha species we examined the stems of Jatropha multifida and isolated two new macrocyclic diterpenoids, 1 and 2 along with ten known compounds. Here, we describe the isolation of these constituents and structure elucidation of the unknown compounds.Compound 1 was isolated as a semi solid. Its molecular formula was deduced to be C 22 H 30 O 5 from its mass spectrum, elemental analysis and 13 C-NMR spectrum. The IR spectrum of the compound indicated the presence of carbonyl group and unsaturation. The 1 H-and 13 C-NMR spectral data of 1 ( Table 1) clearly suggested that its structure is related to that of japodagrone (3) 10) (a known constituent of Jatropha species) which has also presently been isolated from the investigated plant. All of the signals for the protons and carbons in the 1 H-and 13 C-NMR spectra respectively, were assigned from 2D NMR ( 1 H-1 H correlation spectroscopy (COSY), nuclear Overhauser effect spectroscopy (NOESY), heteronuclear single quantum coherence (HSQC) and heteronuclear multiple bonding connectivity (HMBC)) and dis- Chemical investigation on the stems of Jatropha multifida resulted in the isolation of two new macrocyclic diterpenoids whose structures were established by extensive studies of their 1D and 2D-NMR spectra. The occurrence of ten other known compounds in this species is reported here for the first time.
