Inflammation is the response of the body to noxious stimuli such as infections, trauma, or injury. Experimental studies have shown that vanillic acid has antiinflammatory effects. The objective of this study was to investigate the antiinflammatory and antipyretic properties of the derivative of vanillic acid, isopropyl vanillate (ISP-VT), in mice. The results of this study indicated that ISP-VT reduced paw edema induced by carrageenan, dextran sulfate (DEX), compound 48/80, serotonin, bradykinin (BK), histamine (HIST), and prostaglandin E2 (PGE2). Furthermore, ISP-VT reduced recruitment of leukocytes and neutrophils and reduced its adhesion and rolling, and decreased myeloperoxidase enzyme activity (MPO), cytokine levels (tumor necrosis factor-α and interleukin-6), and vascular permeability. ISP-VT also significantly reduced the expression of cyclooxygenase-2 (COX-2) in subplantar tissue of mice. ISP-VT inhibited COX-2 selectively compared to the standard drug. Our results showed that although ISP-VT binds to COX-1, it is less toxic than indomethacin, as evidenced by MPO analysis of gastric tissue. Treatment with the ISP-VT significantly reduced rectal temperature in yeast-induced hyperthermia in mice. Our results showed that the main mechanism ISP-VT-induced anti-inflammatory activity is by inhibition of COX-2. In conclusion, our results indicate that ISP-VT has potential as an anti-inflammatory and antipyretic therapeutic compound. K E Y W O R D Santipyretic, COX-2 inhibition, inflammation, vanillic acid

Summary The structures of two extracts of flavone , obtained from the stem (L40) and leaf (S40) of Labisia pumila (LP), were identified through UHPLC‐MS/MS analysis. An 2,2′‐azobis[2‐methylpropionamidine] dihydrochloride (AAPH)‐induced hemolysis model was used and the results indicate the protective effects of flavone extracts on erythrocytes against AAPH‐induced oxidative stress, in which L40 treatment performed the better. Quantitative analysis of variations in antioxidative‐associated enzyme and non‐enzyme systems within the erythrocytes were conducted and led to the identification of protective pathways in which flavone extracts could pass through the cell membrane and act as AAPH‐free radical scavengers. Absorptivity of flavone extracts was characterised using a Caco‐2 monolayer model, in which the apparent permeability coefficient indicated that both types of flavone extracts were readily absorbed compounds, while L40 showed better intestinal absorptivity. Therefore, we conclude that flavonoid extracts from the leaf of LP show remarkable potential functional effects on human health.

Aims: To evaluate antioxidant, anti-inflammatory and analgesic activities of extracts of Saba senegalensis leaves. Study Design: In vitro antioxidant assay and in vivo anti-inflammatory, analgesic assay of Saba senegalensis extracts. Place and Duration of Study: Saba senegalensis leaves, were collected in the Centre Region of Burkina Faso, in June–July 2015. The experiments were conducted at the department of Medicine and Traditional Pharmacopeia-Pharmacy (MEPHATRA-PH) of Institute of Research in Health Science (IRSS). Methodology: The anti-oedematous tests with carrageenan and the analgesic with acetic acid and investigate effect on isolated organ were carried out. The standards were acetylsalicylic acid and paracetamol. Results: After five hours of carrageenan-induced edema test, aqueous decoction (AD) presented better inhibition on all measure. In fact, at the different doses of 200 mg/kg, 400 mg/kg, and 600 mg/kg it presented percentages of inhibitions respectively of 30.81%, 62.27% and 72.71%. For the analgesic test, the hydroethanol macerate (HEM) showed a better pain reduction compared to the AD with a maximum effect of 77.28% at 400 mg/kg. Antioxidant activity with AD and his fractions shows that AD showed a better activity for the DPPH assay with an IC50 of 1.74 ± 0.10 μg/mL and a reducing power of 59.53 ± 2.16 mmol ET/g Sample. For HEM and his fractions, the ethyl acetate fraction (FHEM-AcOEt) showed a better IC50 of 0.18 ± 0.01 μg/mL for the DPPH test and dichloromethane fraction (FHEM-DCM) a reducing power agent of 88.88 ± 2.65 mmol ET/g Sample. All fractions were endowed with antioxidant properties by both methods. Conclusion: The study findings suggest that the presence of phenolic and terpenoid compounds could explain the antioxidant, anti-inflammatory and analgesic properties of these extracts.
