Anti-Melanogenic Property of Geoditin A in Murine B16 Melanoma Cells

Abstract: Geoditin A, an isomalabaricane triterpene isolated from marine sponge Geodia japonica, has been demonstrated to induce apoptosis in leukemia HL60 cells and human colon HT29 cancer cells through an oxidative stress, a process also interfering with normal melanogenesis in pigment cells. Treatment of murine melanoma B16 cells with geoditin A decreased expression of melanogenic proteins and cell melanogenesis which was aggravated with adenylate cyclase inhibitor SQ22536, indicating melanogenic inhibition was media… Show more

“…The expression of MITF mRNA was actually upregulated in human melanocytes. To further characterize the pathway leading to the CHS‐induced melanin synthesis, we conducted pharmacological inhibitor experiments to verify the involvement of AC or PKA in human skin melanogenesis (Figure F) . The preincubation of melanocytes with a specific AC inhibitor, SQ 22536 (15 μmol/L), resulted in a slight reduction of melanin content.…”

“…The human SCC cell line DJM-1 and mouse melanoma B16 were cultured as described (Seishima et al , 1997 ;Cheung et al , 2012 ) and transient transfection was carried out with the GeneCellin reagent according to the manufacturer ' s instructions (BioCellChallenge, Toulon, France). Immunofl uorescence analysis was conducted as detailed earlier Murase et al , 2012 ) with Alexa Fluor 488-or 568-labeled immunoglobulin G (Molecular Probes, Eugene, Or, USA) as secondary antibodies.…”
