Anti-inflammatory Ingenane Diterpenoids from the Roots of Euphorbia kansui

Abstract: Bioassay-guided fractionation of the ethanolic extract of the roots of led to the isolation of two new ingenane diterpenoids, euphorkans A (1: ) and B (2: ), together with 16 known analogues (3:  - 18: ). Their structures were determined by combined spectral and chemical methods. All the isolates were evaluated for their inhibitory effects on lipopolysaccharide-induced nitric oxide production in RAW264.7 macrophage cells. Compounds 1:  - 6: and 10:  - 13: exhibited pronounced inhibitory activity with IC values… Show more

“…Additionally, 50% ethanolic extract of persimmon leaves potently inhibited the production of NO • , PGE 2 and IL-6 in LPS-induced RAW264.7 cells [32]. Likewise, the ethanolic extract of the Euphorbia kansui root containing ingenane diterpenoids (euphorkans A and B), together with 16 known analogues, exerted an anti-inflammatory activity that was consistent with Q by inhibiting the effects of certain inflammation mediators, such as TNF-α and IL-6, in a concentration-dependent manner through the inhibition of NF-κB activity [33]. Moreover, the ethanolic extract of the QingX-iaoWuWei decoction, containing quercetin and other ingredients, exhibited synergistic anti-inflammatory activity in the LPS-induced RAW264.7 cell culture via JUN, MAPK1 and AKT1 targets and the mitogen-activated protein kinases (MAPK)/phosphatidylinositol-3 kinase (PI3K)/Akt serine/threonine kinases pathways [34].…”

“…Therein, 11 compounds are ingenane-type and jastrophane-type diterpenoids and triterpenoids. Modern pharmacological study showed that kansuiphorin B and kansuinin A that belong to the ingenane-type and jastrophane-type diterpenoids, respectively, could inhibit NF-kB activity to exert anti-inflammatory activity in RAW264.7 macrophage cells based on a bioassay-guided separation [64].…”

“…It can relieve pleural effusion, ascites and urine retention. 1 Besides, modern pharmacology experiments have indicated that Kansui also has attractive pharmacological usage, such as anticancer, 2,3 antivirus, 4 anti-inflammatory, 5 antiallergic 6 and antiasthma 7 activities.…”
