“…The sections were examined with a Leica DMRA microscope for selective incident light fluorescence 8 . For observation of FITC fluorescence (blue narrow‐band excitation) we used an XBO75W lamp and a Chroma high Q filter set containing: (i) exciter filter HQ 480/40; (ii) dichroic beam splitter Q 505 LP; and (iii) emitter filter HQ 535/50.…”

“…The combination of a linear n‐serrated deposition pattern in patient skin and a dermal binding of antibodies on salt‐split skin suggests an antilaminin‐332 mucous membrane pemphigoid or a p200 pemphigoid 7 . To elucidate this further we used knockout skin substrates, lacking either laminin‐332 from a patient with Herlitz junctional epidermolysis bullosa, or lacking type VII collagen from a patient with recessive dystrophic epidermolysis bullosa 8 . Both substrates were incubated with patient serum.…”

“…Tests on skin deficient in type VII collagen gave negative or reduced staining in 8/14 group I patients, hence confirming the presence of collagen VII autoantibodies ( Fig. If positive BMZ reactions are seen on normal skin by IIF, further tests on collagen VII deficient substrates can help differentiate between these diseases [3]. 7/8 of these patients had ELISA unit values greater than the cut off value (median 82.4 U/ml).…”
