Annexin A5-Conjugated Quantum Dots with a Paramagnetic Lipidic Coating for the Multimodal Detection of Apoptotic Cells

Abstract: Apoptosis, or programmed cell death, plays an important role in the etiology of a variety of diseases, including cancer. Visualization of apoptosis would allow both early detection of therapy efficiency and evaluation of disease progression. To that aim we developed a novel annexin A5-conjugated bimodal nanoparticle. The nanoparticle is composed of a quantum dot that is encapsulated in a paramagnetic micelle to enable its use both for optical imaging and MRI. Multiple recombinant human annexin A5 protein molec… Show more

“…11 Lipid-based nanoparticles, such as liposomes, micelles, and microemulsions, have been evaluated as carriers of MRI contrast and fluorescent agents for noninvasive imaging. 9,[12][13][14][15][16][17][18][19][20] There are several recent reviews on lipid-based paramagnetic nanoparticles 21,22 and on paramagnetic and fluorescent nanoparticles. 3,10,23,24 Kostarelos and Miller described a modular design for targeted paramagnetic liposomes carrying nucleic acids, termed Gd-ABCD.…”

“…The fluorophores involved in these bioconjugates, however, were not QDs but fluorescein isothiocyanate (FITC), which might limit the applications for these bioconjugates because of the disadvantages of this organic dye. A hallmark of apoptosis is the externalization of phosphatidylserine, which is recognized by annexin V. Accordingly, van Tilborg et al (35 ) and Prinzen et al (36 ) used annexin V-conjugated QDs coated with a paramagnetic lipid to detect apoptotic cells with fluorescence and magnetic resonance imaging. We recently demonstrated that trifunctional avidin-coupled nanospheres are capable of recognizing apoptotic cells preincubated with annexin V-biotin via the strong biotin-avidin interaction (31 ).…”
