Angiotensin II Activation of Insulin-Like Growth Factor 1 Receptor Transcription Is Mediated by a Tyrosine Kinase–Dependent Redox-Sensitive Mechanism

Abstract: Abstract-We have recently shown that angiotensin II activation of insulin-like growth factor 1 receptor transcription is a critical requirement for angiotensin-stimulated vascular smooth muscle cell growth; therefore, we examined the signaling pathway involved. In rat aortic smooth muscle cells, the antioxidants N-acetyl-L-cysteine (5 mmol/L) and pyrrolidine dithiocarbamate (100 mol/L) completely inhibited angiotensin II-stimulated increases in IGF-1R mRNA and protein levels, suggesting the involvement of rea… Show more

“…ANG II binding to AT 1 R leads to the redox-sensitive transactivation of endothelial growth factor receptor (413) and platelet-derived growth factor receptor (349). ANG II also transactivates the insulin-like growth factor (IGF) receptor (IGFR), at least in part by a redox-sensitive mechanism (119).…”

“…Regarding the relation between the RAS and IGF signaling, early work showed that ANG II induces vascular smooth muscle cell mitogenesis by increasing the number of IGFR and upregulating IGF1R mRNA (109,119). Later, stimulation of the cardiac and systemic IGF-I pathways was found to make a chief contribution to ANG II-induced cardiac remodeling (42).…”

“…In vascular smooth muscle, angiotensin II stimulates cell growth through activation of IGF1R transcription. This effect is mediated via calcium-dependent activation of protein tyrosine kinase activity (Du et al 1999). We found downregulation of IGF1R and CALM1, the latter involved in calcium signalling, which indicates that the IGF1 signalling pathway may be a mediator of the effects of angiotensin II on the proliferation of adrenocortical cells.…”

“…For example, exposure of VSMCs to ROS, either by H 2 O 2 or ox-LDL (oxidized-low-density lipoprotein), has been reported to either increase [12] or decrease [13] IGF1R expression. The molecular and transcriptional mechanisms of IGF1R expression in response to oxidative stress have not been examined.…”

“…IGF1R-dependent signalling is crucial for the survival of many cell types, including VSMCs [9,10]. IGF1R activity in VSMCs is mediated by a number of intracellular and extracellular factors [11][12][13][14], and its expression is also regulated at the level of transcription [15]. However, the role of oxidative stress on IGF1R gene expression in VSMCs is not clear.…”
