Angelica polysaccharide ameliorates memory impairment in Alzheimer’s disease rat through activating BDNF/TrkB/CREB pathway

Abstract: This study aimed to investigate the effect of Angelica sinensis polysaccharides (ASP) on Alzheimer’s disease (AD) and its underlying mechanisms. In our study, we build the AD model by injecting Aβ25–35. Morris water maze (MWM) was applied to investigate learning and memory. Moreover, neurotransmitters, free radical, and inflammatory factors were also measured. Pathological change and neuronal death in hippocampus CA1, CA3, and DG region were detected by HE staining and Nissl staining. The neuronal apoptosis wa… Show more

“…ASP, as one of the main polysaccharides extracted from the Angelica root, has anti-tumor and anti-inflammatory activities. At the same time, ASP has a high affinity for ASGPR and MR of hepatoma carcinoma cells (Zhang et al., 2018 ; Du et al., 2020 ), so it is expected to become a natural product for the preparation of liver-targeted carrier materials (Du et al., 2020 ). In this paper, an ASP-based nanocarrier was prepared by a self-assembling method in order to achieve liver targeting.…”

“…After the rats were injected intraperitoneally with 10% chloral hydrate, aggregated Aβ 25-35 was then injected into the bilateral hippocampus by a stereotaxic apparatus to construct the AD rat model. Morris water maze (MWM) was applied to evaluate the spatial learning and memory ability of rats [ 26 ]. MWM test is mainly divided into location and navigation test and space exploration test.…”

“…These functions of BDNF/TrkB signaling are achieved by a combination of three downstream signaling cascades: the phosphatidylinositol 3-kinase (PI3K), mitogen-activated protein kinase (MAPK) and phospholipase-C gamma (PLCγ) pathways (Edelbrock et al, 2018). Moreover, the activation of BDNF/TrkB signaling results in the phosphorylation of the transcription factor CREB (Shang et al, 2019;Du et al, 2020). Blocking BDNF signaling leads to reduced CREB transcription, which is essential for synaptic plasticity and learning and memory (Mowla et al, 1999).…”
