Inflammation is a complex biological response to stimulation. Natural cassane diterpenoids from Caesalpinia genus exhibit significant anti-inflammation activity. Eight new cassane diterpenoids (1-8) along with seven known ones (9-15) were obtained from the seed kernels of Caesalpinia cucullata Roxb. This is the first report on chemical investigation of the seed kernels of C. cucullata, and the cassane diterpenes were found in this plant for the first time. Their structures were elucidated based on the extensive spectroscopic analyses, and the absolute configurations were identified by ECD calculation and X-ray crystallography. All compounds were evaluated for their anti-inflammation activity by inhibiting NO production in LPS-induced RAW 264.7 cells. Compounds 1-2 and 9-11 exhibited effective inhibitory activity with inhibition rate more than 50%. The iNOS enzyme activity and molecular docking experiments were performed to explore the preliminary mechanism. Eventually, a potential anti-inflammatory mechanism revealed that the bioactive cassane inhibited overproduction of NO by targeting key residues in the iNOS active cavity to reduce iNOS enzymatic activation.

Network Pharmacology-Based Analysis on Lonicera japonica for Chronic Osteomyelitis Treatment

Ethnopharmacological Relevance. Lonicera japonica (LJP) is a broadly used traditional Chinese medication treatment for chronic osteomyelitis (COM). But, the main antiosteomyelitis compounds and functional targets of LJP are still unclear. Aim of the Study. To screen LJP drug targets and active compounds in COM treatment. Materials and Methods. Active compounds of LJP were examined established on the analysis platform, Traditional Chinese Medicine Systems Pharmacology (TCMSP) database. DrugBank identified drug targets and annotated them on UniPort and GeneCards. Besides, the COM-related genes were identified on GeneCards. The network of the drug, main active compounds, targets, and diseases was built utilizing Cytoscape. STRING was utilized to build the protein-protein interaction network. Moreover, the KEGG and GO pathway enrichment analysis were applied to analyze biological function. Results. 23 active compounds of LJP were screened, and 204 drug targets and 686 COM-related genes were identified. Forty-five intersection genes were overlapped from 204 drug targets and 686 COM-related genes. The drug-active compounds-target protein-diseases network was established based on 23 active compounds of LJP and 45 intersection genes. Moreover, the interaction of 45 intersection genes was explored by the PPI network, and the drug-active compounds-target protein-diseases network was formed grounded by 23 active compounds of LJP, 45 intersection genes, and PPI network. The KEGG and GO pathway enrichment analysis specified that 45 intersection genes primarily enriched in immune-related pathways and oxidative stress-related pathways. Conclusions. In the research done, the main active compounds of LJP and drug targets in the treatment of COM were identified. Our findings might provide the ingredient option of LJP and drug targets of LJP in COM treatment.

Background and Purpose Neutrophils and the release of neutrophil extracellular traps (NETs) play important roles in the pathogenesis of RA. However, IgD, which was abnormally higher in RA, has not been studied for its pathological role in neutrophil activation and NETs formation. Experimental Approach Peripheral blood of RA patients and healthy controls were collected and adjuvant-induced arthritis (AA) rat models were established. Body weights, the severity of arthritis of AA rats were monitored regularly. After being stimulated with IgD, expression of FcδR on neutrophils and NETs formation were analyzed with multiple approaches such as flow cytometry, scanning electron microscopy, western blot, and qPCR. IgD-Fc-Ig were used to block interactions between IgD-FcδR. Additionally, the effect of IgD-induced neutrophils or NETs on FLS was assayed. Key Results As a specific marker of NETs, the level of citrullinated histone H3 was positively correlated with sIgD and FcδR in RA patients. IgD enhances the release of NETs by activating neutrophils. IgD-Fc-Ig could significantly reduce NETs formation and FcδR expression on neutrophils in vitro. In vivo, IgD-Fc-Ig treatment significantly regulates the neutrophil activity and NETs formation. IgD-Fc-Ig could restrain IgD-induced neutrophil activation and NETs formation, thus inhibiting FLS proliferation. Conclusions and Implications Neutrophils were activated by IgD, which suggests that neutrophils play a role in inducing FLS proliferation in RA patients who have abnormally higher IgD levels, there by increasing the severity of the disease. Blocking FcδR inhibited neutrophil activation and may represent an additional attractive novel therapeutic strategy for the treatment of RA.
