Analysis of the levels of Th9 cells and cytokines in the peripheral blood of mice with bronchial asthma

Abstract: The purpose of the study was to detect the level of T-helper type 9 (Th9) cells and the cytokine interleukin-9 (IL-9) in peripheral blood of mice with bronchial asthma, and to explore the relationship between the expression of Th9 cells and the pathogenesis of asthma. Thirty female-specific pathogen-free (SPF) Bagg' albino (BALB)/c mice were selected and randomly divided into the control group (n=15) and the bronchial observation group (n=15). Mice in the bronchial observation group were treated with ovalbumin… Show more

“…Jia et al(found that the levels of IL-9 and IL-4 in peripheral blood of asthmatic children were significantly higher than those of healthy children. In addition, animal experiments showed that not only the proportion of Th9 cells in peripheral blood and the level of IL-9 in asthma model mice were significantly higher than those in control group, but also the level of IL-9 mRNA in lung tissue was significantly increased (20). Our study observed IL-9 level in lung tissue and BALF and the proportion of CD4 + Th9 cells in the PBMCs of asthma group were significantly higher than that of control group, which was consistent with the existing reporters.…”

“…Hereby, the study shows that during AAI TGF-bsignaling in CD4 + T cells is an important factor for allergogenic Th2, Th9 and Th17 cell activity in vivo, while the Treg compartment remained largely unaffected. Th2, Th9 and Th17 cell recruitment upon allergen challenge (70) was reduced in absence of TGFBR2 along with respective transcription factors PU.1 (Th9), GATA3 (Th2), and NFAT5. NFAT5 is an osmosensitive transcription factor that negatively regulates IL-17 and IFN-g gene expression (71) and enhances Th2 response (72).…”

“…Hereby, the study shows that during AAI TGF-β-signaling in CD4 + T cells is an important factor for Th9 cell differentiation in vivo, while the Treg compartment remained largely unaffected. Th2, Th9 and Th17 cell recruitment upon allergen challenge 39 was reduced in absence of TGFBR2 along with respective transcription factors PU.1 (Th9) and GATA3 (Th2). Moreover, the transcription factor FOXO1, which was recently described to be essential for IL-9 production and Th9 cell differentiation, thus promoting airway allergy 25, 40, 41 , was clearly reduced upon TGFBR2-ablation.…”
