Analysis of the Association of Matrix Metalloproteinase-1 Gene Promoter (rs1799750) Polymorphism and Risk of Ovarian Cancer

Abstract: In summary, this meta-analysis showed that the MMP1 1607-bp polymorphism is not associated with ovarian cancer risk.

“…A total of 17 articles [ 11 – 27 ] were identified through search strategy. Reviewed on abstracts among these articles, 4 articles were excluded because 3 articles [ 25 – 27 ] were meta-analysis and 1 article [ 13 ] could not present detailed data. Then 13 full text articles were obtained for further evaluation, in which 3 articles were deleted for 2 articles [ 16 , 21 ] were duplicated publication and 1 article [ 17 ] had no control group.…”

“…Moreover, the 2G allele of MMP1 rs1799750 is associated with the risk of several types of cancer, including oral (12), breast (13), gastric (18), colorectal (26) and bladder (27). In addition, a negative association was also found in ovarian (14) and lung (15) cancer. However, little is known about the association between MMP1 genotype and the risk of leukemia, which is a nonsolid type of cancer.…”

“…MMP1, also known as collagenase-1, is most abundant among the MMPs and under the control of activator protein-1 (AP1), which binds to the promoter region of mitogen-activated kinase through polyomavirus-enhancing activity-3 (10,11). In 2016, a polymorphic variation was found in the MMP1 promoter region at upstream position 1,607 bp (rs1799750, also known as rs11292517, rs17886084, rs61633513, rs139258005, rs375359915 and rs368625565), which controls the transcriptional activity of MMP1 and was associated with the incidence and progression of several types of cancer, including oral (12), breast (13), ovarian (14), lung (15), esophageal (16,17), gastric (18) and colorectal (19).…”

“…In order to increase the sensitivity and specificity for the detection of EOC, a number of novel markers have been investigated, including matrix metalloproteinase-1, osteopontin, and human epididimis 4 [4][5][6]. Of these, high mobility group box 1 (HMGB1) was characterized as a non-histone, nuclear DNA-binding protein [7].…”

“…However, its utility as a screening marker is limited due to its low sensitivity and specificity [2]. Moreover, the serum level of CA125 can be elevated in other cancers such as uterine cancer, gastrointestinal cancer, as well as in benign disease [3].In order to increase the sensitivity and specificity for the detection of EOC, a number of novel markers have been investigated, including matrix metalloproteinase-1, osteopontin, and human epididimis 4 [4][5][6].
