Analysis of Proinflammatory and Anti-Inflammatory Cytokine Serum Concentrations in Patients With Multiple Sclerosis by Using a Multiplexed Immunoassay

Abstract: We examined cytokines and other inflammatory markers in serum samples from 833 patients with multiple sclerosis and 117 healthy control subjects. A multiplexed immunoassay was used to assess the concentrations of 13 cytokines/inflammatory markers: interferon (IFN)-γ; interleukins (ILs)-1β, 2, 4, 5, 6, 8, 10, 12, and 13; tumor necrosis factor (TNF)-α; IL-2 receptor; and soluble CD40 ligand. Significant increases between patients and control subjects were found for IFN-γ (mean, 7.5 vs 0.4 pg/mL; P = .0002), IL-2… Show more

“…These data are supportive of the possible involvement of TMEM66 in the pathological events of MS in the patients, as MS is considered to be mainly mediated by immune response (31). The proinflammatory and anti-inflammatory responses were shown to have clear effects over the time in the progression of the disease (12).…”

“…Such a disruption might play a role in the increased incidence of various medical conditions in shift workers, such as the increased risk for infections, autoimmune diseases, cancer, and cardiovascular and metabolic disorders (2,(8)(9)(10)(11)(12)(13). Interestingly, an increased prevalence of autoimmune conditions, such as multiple sclerosis and autoimmune thyroid disorders, was associated with shift work (11,12,(60)(61)(62)(63). In autoimmune thyroid disorders, there is an increase in the recruitment of Th1 lymphocytes in thyroid tissue (based on changes in cell trafficking between blood and tissues), leading to an increase in the production of IFN-g and TNF-a, further stimulating the recruitment of Th1 lymphocytes and initiating an amplification feedback loop (63).…”

“…Experimental autoimmune encephalomyelitis (EAE) is an acceptable model for the study of MS which is induced by immunization with myelin antigens or by adoptive transfer of myelin-specifi c T cells (2). Investigating changes in proinfl ammatory and anti-infl ammatory cytokines over time of disease progression have led to an insight into the pathogenesis of MS (3).…”
