Analysis of coagulation factor IX in bioreactor cell culture medium predicts yield and quality of the purified product

Abstract: Coagulation factor IX (FIX) is a highly complex post-translationally modified human serum glycoprotein and a high-value biopharmaceutical. The quality of recombinant FIX (rFIX), especially complete g-carboxylation, is critical for rFIX clinical efficacy. Changes in bioreactor operating conditions can impact rFIX production and occupancy and structure of rFIX posttranslational modifications (PTMs). We hypothesized that monitoring the bioreactor cell culture supernatant with Data Independent Acquisition Mass Spe… Show more

“…The top 25 mL of supernatant were transferred to 3 kDa Amicon columns (Amicon Ultra-15 Centrifugal Filter Unit, UFC 900324) and concentrated following the manufacturer’s recommendations. 100 μL of the concentrated sample were precipitated overnight in 4 volumes of methanol:acetone in a LoBind Eppendorf tube, as previously described ( Zacchi et al, 2020a , 2020b ). The supernatant was eliminated after high speed centrifugation, and air-dried proteins were resuspended in 50 mM ammonium bicarbonate supplemented with 10 mM DTT and 1 μg of trypsin.…”
