Analysis of association of clinical aspects and IL1B tagSNPs with severe preeclampsia

Abstract: We identified an association between clinical variables and preeclampsia. Univariate analysis suggested that inflammatory process-related genes, such as IL1B, may be involved and should be targeted in further studies. The identification of the genetic background involved in preeclampsia host response modulation is mandatory in order to understand the preeclampsia process.

“…IL-1β gene polymorphisms have been associated with preeclampsia 62,63 , preterm birth 75 , and recurrent pregnancy loss 76 . In their Brazilian study, Leme et al 77 , have found an association of preeclampsia risk with the IL-1β rs1143630 T allele, while Pontillo et al 78 , who also conducted their study in Brazil, have found no association of IL1β rs1143634 with preeclampsia risk, a finding similar to that of other studies 79 . The -511 C/T polymorphism in the IL-1β gene promoter region is implicated in the differential production of cytokines.…”

“…A possible association between SNPs in IL1B was investigated in Brazilian women with severe PE. In this study, the “rs1143630 T” allele was associated with PE (Leme Galvão et al, 2016). In a Maya-Mestizo population sample, no association between TNFA (−308G/A, −850C/T) SNPs and PE was observed (Canto-Cetina et al, 2007).…”

“…These opposing signals may provide an explanation for the variation in fetal outcomes observed in pregnancies complicated by obesity. IL-1β appears to be consistently associated with hypertensive disorders such as preeclampsia (Kalinderis et al 2011; Dong and Yin 2014; Leme Galvão et al 2016) and may be predictive of future disease in early to mid pregnancy (Siljee et al 2013; Taylor et al 2016). Enhanced placental secretion may be the primary source of circulating of IL-1β (Amash et al 2012).…”
