Cardiac Oxidative Status in Diabetic Wistar Rats Exposed To Ethanol Extract of Cucumis Sativus Fruit

The present study investigated cardiac oxidative status in diabetic Wistar rats exposed to ethanol extract of Cucumis sativus fruit. Male Wistar albino rats (n = 25, mean weight = 215 ± 15 g) were randomly assigned to five groups of 5 rats each: normal control, diabetic control, metformin, extract (200 mg/kg body weight, bwt) and extract (300 mg/kg bwt) groups. Diabetes mellitus was induced in the rats via intraperitoneal injection of 50 mg/kg bwt STZ. The diabetic rats were treated for 21 days with either metformin (50 mg/kg bwt) or the extract. Activities of antioxidant enzymes such as catalase, superoxide dismutase (SOD), glutathione peroxidase (GPx) and glutathione reductase (GR) as well as molecules like reduced glutathione (GSH), total protein (TP), malondialdehyde (MDA) and nitric oxide (NO) were measured in heart homogenate. The results showed that induction of diabetes mellitus with STZ significantly increased the fasting blood glucose (FBG) concentrations of the rats, while decreasing the activity/concentration of antioxidant enzymes/ molecules (p < 0.05). However, treatment of the diabetic Wistar rats with the extract markedly reduced the FBG concentration and body weights of rats, but enhanced the activity/concentration of antioxidant enzymes/molecules in cardiac tissue (p < 0.05). These results indicate that ethanol extract of the medicinal plant fruit has the potential to promote antioxidant defense in the heart of STZ-induced diabetic rats.

The present study investigated the effect of methanol fraction of ethanol extract of Dialium guineense (MEDG) stem bark on streptozotocin (STZ)-induced oxidative stress in rat liver. Male albino rats of Wistar strain (n = 25, mean weight = 215 ± 15 g) were randomly assigned to five groups (5 rats/group): normal control, diabetic control, metformin, MEDG [200 mg/kg body weight (bwt)] and MEDG (300 mg/kg bwt) groups. Diabetes mellitus was induced in the rats via intraperitoneal injection of 50 mg/kg bwt STZ. The diabetic rats were treated for 21 days with either metformin (50 mg/kg bwt) or the extract at doses of 200 and 300 mg/kg bwt, respectively. Activities of antioxidant enzymes such as catalase, superoxide dismutase (SOD), glutathione peroxidase (GPx) and glutathione reductase (GR) as well as antioxidant vitamins (retinoic acid, ascorbic acid and α-tocopherol) were measured in 20 % liver homogenate. The results showed that induction of diabetes mellitus with STZ significantly increased the fasting blood glucose (FBG) concentrations of the rats (p < 0.05). However, treatment of the diabetic rats with the extract markedly reduced the blood glucose concentration and body weights of rats (p < 0.05). Treatment of diabetic Wistar rats with MEDG stem bark significantly increased the activities of the antioxidant enzymes and molecules as well as concentrations of nitric oxide (NO), but it markedly reduced the concentrations of hepatic total protein (TP) and malondialdehyde (MDA) (p < 0.05). These results indicate that MEDG stem bark can potentiate antioxidant defense in diabetic rat liver.

The world population is expected to double by 2050, creating a greater demand for animal protein sources. This calls for an increase in the supply of animal-derived foods in order to avoid food scarcity in the future. Non-ruminants, which would have been a panacea to this increased demand for animal-derived foods, have suffered from the rising costs of feed ingredient world-wide. The increase in the prices of conventional feedstuffs have attracted the attention of researchers on the potentials of lesser-known tropical medicinal plants like Dialium guineense Willd (black velvet tamarind) as a feed resource for non-ruminants. The increase in prices of conventional feedstuffs have attracted the attention of researchers on the potentials of lesser-known tropical medicinal plants. One such lesser-known tropical plant that can be used in chicken and livestock production is black velvet tamarind. The plant is found in many parts of the world. It belongs to the family Fabaceae, has high forage yield, resistant to drought and insects, and does not require annual cultivation. The whole seed is moderate in crude protein and fibre but high in carbohydrates. The leaf, on the other hand, is low in carbohydrate, moderate in crude protein, and high in ash. D. guineense stem bark and fruit are excellent source of essential oils, and rich source of dietary fibre, minerals, and vitamins for monogastrics. The plant is endowed with beneficial phytochemicals such as alkaloids, tannins, phenols, flavonoids which may serve as natural alternatives to in-feed antibiotics. D. guineense has beneficial pharmacological effects including antioxidant and antimicrobial properties. The purpose of this study was to review the body of knowledge on the nutritional and chemical composition of D. guineense and its impact in animal production.
