Our findings were different than those of Saino et al (1976) [49], who suggested that it was possible to predict that an incremental increase in the amount of P. acnes could lead to worse acne lesions. Even in Groups 1 and 3 (which were treated with lymecycline), an increase in the C15:0 levels from D30 to D90 ( P = 0.006) was observed, which could indicate that pentadecanoic acid is not an important constitutional element for this bacteria or that it is, in fact, a marker of unfeasible (devitalized) bacteria.…”

Background. Acne is a dermatosis that involves an altered sebum pattern. Objectives. (1) To evaluate if a treatment based on antibiotics (lymecycline) can alter fatty acids contents of the sebum of patients with acne; (2) to evaluate if oral supplementation of fatty acids can interfere with fatty acids contents of the sebum of patients with acne; (3) to evaluate if there is any interaction in fatty acids contents of the sebum of patients with acne when they use both antibiotics and oral supplementation of fatty acids. Methods. Forty-five male volunteers with inflammatory acne vulgaris were treated with 300 mg of lymecycline per day, with 540 mg of γ-linolenic acid, 1,200 mg of linoleic acid, and 510 mg of oleic acid per day, or with both regimens for 90 days. Every 30 days, a sample of sebum from the forehead was collected for fatty acids' chromatographic analysis. Results. Twelve fatty acids studied exhibited some kind of pattern changes during the study: C12:0, C14:0, C15:0, C16:1, C18:0, C18:1n9c+C18:1n9t, C18:2n6t, C18:3n6, C18:3n3, C20:1, C22:0, and C24:0. Conclusions. The daily administration of lymecycline and/or specific fatty acids may slightly influence some fatty acids levels present in the sebum of patients with inflammatory acne vulgaris.

Biphasic protection against bacterial infection in mice induced by vaccination of Propionibacterium acnes

A single intraperitoneal injection of the phenol-treated cells of Propionibacterium acnes into mice showed nonspecific resistance against subsequent lethal doses of an intraperitoneal challenge of Klebsiella pneumoniae, Staphylococcus aureus, and Streptococcus pyogenes. The protection showed a biphasic pattern. The maximum protection, designated as the early phase protection, was seen in mice injected with P. acnes vaccine 1 to 2 days before the challenge, whereas the late phase protection was seen in mice vaccinated 16 to 22 days before the challenge. The activity of the reticuloendothelial system in mice after vaccination also showed a biphasic pattern with the peak on days 4 and 12. The delayed activation of the reticuloendothelial system lasted up to 3 weeks and coincided with the period of the late phase protection. The early phase resistance was markedly impaired by the treatment with hydrocortisone and carrageenan, but not by the treatment with anti-thymocyte serum, actinomycin D, or cyclophosphamide. The number of peritoneal polymorphonuclear leukocytes in vaccinated mice increased on days 1 to 2. The number of macrophages also increased at 2 to 21 days after vaccination and reached its maximum on day 14. Total activities of acid phosphatase, Nitro Blue Tetrazolium reduction, and the phagocytic activities of peritoneal exudate cells were also enhanced on and after day 1 after the injection of P. acnes vaccine.

Biochemical and serological characteristics of 128 strains of Propionibacterium acnes isolated from the facial skin of healthy Japanese volunteers were compared with the three standard strains of the American Type Culture Collection, ATCC 6919,11827, and 11828. Accordingly, the isolated strains ofP. acnes were classified into five biotypes (B1 to B5) on the basis offermentation tests of ribose, erythritol, and sorbitol. Two serotypes were distinguished by the agglutination test. P. acnes belonging to serotype I had galactose as a cell wall sugar, whereas those of serotype II lacked galactose. The strains of serotype I were distributed among all five biotypes (Bi to B5); however, those of serotype II consisted only of one biotype (B2). A term "sero-biotype" was introduced to differentiate and carefully classify the isolates. The predominant sero-biotypes differed with the individual and region of the facial skin. In general, strains of sero-biotype IB1, IB3, IB4, and IIB2 were more frequently isolated than those of sero-biotype IB2 and IB5. Thus, for routine assay work, serotyping of P. acnes as based on erythritol and sorbitol fermentation is both practical and applicable.
