An Inexpensive and Simple Method for Preparing Chromosome Spreads

Abstract: 717for the metabolism of PCPE. If this is true, our experiments raise the questions as to whether the substance excreted in the urine and characterized as "pregnanediol" by the Klopper method is actually 5P-pregnan-3~, 2 0 a-diol or alternatively, whether the "pregnanediol" measured originates from PCPE or arises from endogenous sources under the influence of PCPE. To answer these points, the metabolism of isotopically labeled PCPE must be studied in animals as well as in humans.Summary. Progesterone and its 3… Show more

“…Chromosome medium la (Gibco Bio-Cult) was used for the cultures (0.5 ml blood/3.5 ml medium). This medium is an unpublished Gibco modification of the medium developed by Razavi (1965), containing Eagle's minimal essential me dium (MEM) for suspension cultures, fetal calf serum, heparin, antibiotics, and phytohemaggluti nin. Distamycin A HC1 (Boehringer, Mannheim) was dissolved in distilled H.,0 (4 mg/ml), sterilized by filtration, and added to half of the cultures of each individual 24 h before fixation at a final concentration of 100 «g ml.…”

“…The number of colonies on the surface of the spleen were counted as described by T ill and M cC ulloch [22], Erythropoie tin was a LD fraction obtained from the filtrate of anemic rabbit plasma [14] and its potency tested against a sample of the International Reference Preparation [3]. PHA was an extract of Phaseolus vulgaris prepared according to the technique de scribed by Razavi [16]. In pilot experiments it was found that spleen weight in creases proportionally to the increase in the dose range from 0.05 to 0.30 ml.…”
