The purpose of this study was to compare the antibacterial properties of Azadirachta indica (neem) or Curcuma longa (turmeric) against Enterococcus faecalis with those of 5% sodium hypochlorite or 2% chlorhexidine as root canal irrigants in vitro. The activity of neem, chlorhexidine, sodium hypochlorite, or turmeric against E. faecalis was measured on agar plates using the agar diffusion method. The tube dilution method was used to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the irrigants used. Chlorhexidine or neem exhibited the greatest antibacterial activity when used as endodontic irrigants against E. faecalis, followed by sodium hypochlorite. No statistically significant difference was observed between neem, sodium hypochlorite, or chlorhexidine. The MIC of neem was 1: 128, which was similar to that of chlorhexidine. The MBC for each of these irrigants was 1: 16. Neem yielded antibacterial activity equivalent to 2% chlorhexidine or sodium hypochlorite against E. faecalis, suggesting that it offers a promising alternative to the other root canal irrigants tested.

The study was conducted using the agar diffusion test. The medicaments used were chlorhexidine gel, calcium hydroxide, and a mixture of chlorhexidine and calcium hydroxide. Enterococcus faecalis and Candida albicans microorganisms were used. A total of six petri dishes were prepared for each microorganism to be three units. Three wells were prepared on each petri dish, 5 mm in diameter and 2 mm deep, with the agar medium. The freshly prepared medicaments were placed on the agar plates, which were inoculated with the test microorganisms. After incubation periods of 24, 48, and 72-hours, the zones of the growth inhibition were observed and measured. Data were statistically evaluated. The chlorhexidin (CHX) gel group showed significantly lager inhibition zones for both E. faecalis and C. albicans in the 48-hours time period, when compared to the other experimental groups (p <0.001). Disinfection and shaping of the canal with a combination of chemical agents and endodontic instruments play important roles in the success of endodontic therapy. Root canal irrigants aid in achieving disinfection in canal systems inaccessible to biomechanical preparation. A wide variety of synthetic irrigants are available today but some may be ineffective, and there may be safety concerns and side effects. Herbal alternatives may be advantageous. To evaluate the antibacterial properties of herbal extracts, namely turmeric and triphala as irrigants during endodontic treatment in comparison with the conventional irrigating solutions. Agar well diffusion assay and MIC methods were used to determine the efficacy of the experimental irrigant in removing E. faecalis. The difference between groups was statistically analysed. 2% chlorhexidine showed the highest zone of inhibition against E. faecalis followed by triphala. There was no significant difference between triphala and turmeric but triphala showed more inhibition effect against E. faecalis because of chelating properties. Chlorhexidine has significant antimicrobial efficacy against E. faecalis. Considerable reduction in growth of E. faecalis was seen in the herbal extract groups and considering their non-toxic nature and other physiological benefits, further studies are warranted. Herbal irrigants, E. faecalis, chlorhexidine. The goal of endodontic therapy is the removal of all vital or necrotic tissue, microorganisms, and microbial by-products from the root canal system. This may be achieved through chemo-mechanical debridement of the root canal. 1 The complex nature of the root canal complex and the presence of many inaccessible areas requires a combination of mechanical instrumentation and irrigation to decrease the amount of bacteria/microorganisms in the system. 2 E. faecalis is an enteric facultative gram positive bacterium which can grow independently in the root canal. 2 It is the most commonly isolated species from the root canals of the teeth, especially those with failed endodontic treatment. 3,4
