An immunochromatographic assay for rapid etection of salvinorin A

Abstract: We developed an immunochromatographic assay (ICA) that enables rapid analysis of salvinorin A (Sal A) in Salvia divinorum within 10 min. The result shows that no Sal A in other samples of Lamiaceae plants was detected, but it could recognize Sal A among other substances in complex samples. The main advantage of the ICA is its high performance in combination with low cost, simplicity, and speed. Our newly developed combined ICA/indirect competitive ELISA(icELISA) system enables analysis of large numbers of samp… Show more

“…The compounds with carboxylic acid and hydroxyl functional groups are usually conjugated by a carbodiimidemediated reaction [33,34,47,[95][96][97][98][99][100][101][102], whereas compounds with active hydrogen, e.g., α-picolines, ketones, esters, and acetylenes, can be conjugated to an amine or amide group of the carrier by a Mannich reaction [103,104]. Furthermore, structure modifications of the parent compounds to add the reactive functional group to the molecule can be conducted when necessary.…”

“…Salvinorin A acts as a selective agonist for Kappa opioid receptors (KOR), thereby exerting its potent hallucinogenic effects [ 12 , 142 ]. A dose of between 200 μg and 500 μg is capable of inducing deep hallucinations with extraordinary illusions and a feeling of physical and mental displacement [ 143 , 144 ]. However, some studies conducted with S. divinorum and its bioactive compound salvinorin A have shown that it has some effects with therapeutic potential, such as drug addiction, pain treatment, neurological, gastrointestinal diseases and anti-inflammatory agent [ 145 , 146 , 147 , 148 , 149 , 150 , 151 , 152 , 153 , 154 , 155 , 156 ].…”

“…Several methanol extracts of S. divinorum and other species of the Lamiaceae family were analysed with the immunochemical method developed, with salvinorin A being only detected in S. divinorum, at concentrations varying from 9.72 to 16.7 µg/mg dry wt [94]. A few years later, the research group developed a new immunochromatographic assay (ICA) using the aforementioned monoclonal antibody combined with the icELISA, aiming at identifying salvinorins in S. divinorum [95]. The combined technique resulted in higher sensitivity, accuracy, and reliability of detection.…”
