An Nfic-hedgehog signaling cascade regulates tooth root development

Abstract: Coordination between the Hertwig's epithelial root sheath (HERS) and apical papilla (AP) is crucial for proper tooth root development. The hedgehog (Hh) signaling pathway and Nfic are both involved in tooth root development; however, their relationship has yet to be elucidated. Here, we establish a timecourse of mouse molar root development by histological staining of sections, and we demonstrate that Hh signaling is active before and during root development in the AP and HERS using Gli1 reporter mice. The pro… Show more

“…On the other hand, inhibition of the Shh pathway before molar root initiation, via treatment with Hh inhibitor, also leads to decreased mesenchymal cell proliferation around the HERS and reduced root length . Taken together, these data suggest a dose-dependent role for Shh signaling activity in the regulation of molar root development Liu et al, 2015). As mentioned above, Nfic interacts with Tgfβ signaling during root development, but it is also an important downstream target of Gli1-mediated Hh signaling in the root (Huang et al, 2010).…”

“…Mutations in DLX3 are also associated with amelogenesis imperfecta hypoplastichypomaturation with taurodontism in humans (Dong et al, 2005). Of note, the ablation of Smad4, Nfic or Wnt10a in mice leads to similar phenotypes Liu et al, 2015;Yang et al, 2015).…”

“…Nuclear factor I‐C (NFIC) has long been believed to play an essential role in tooth formation. Disruption of the Nfic gene in mice leads to abnormal tooth roots that are predominantly caused by abnormal odontoblast differentiation and dentin formation during tooth formation (15, 16).…”

“…BMPs in the dental mesenchyme may participate in the formation of the HERS because BMPs are expressed strongly before root development . NFI‐C has been shown as an important factor for root elongation as Nfic KO mice display no root formation . Here, we observed no difference in the Nfic expression between Bmpr1a cKO and control molars.…”
