An. gambiae gSG6-P1 evaluation as a proxy for human-vector contact in the Americas: a pilot study

Abstract: BackgroundDuring blood meal, the female mosquito injects saliva able to elicit an immune response in the vertebrate. This immune response has been proven to reflect the intensity of exposure to mosquito bites and risk of infection for vector transmitted pathogens such as malaria. The peptide gSG6-P1 of An. gambiae saliva has been demonstrated to be antigenic and highly specific to Anopheles as a genus. However, the applicability of gSG6-P1 to measure exposure to different Anopheles species endemic in the Ameri… Show more

“…Synthetic peptides were already used to assess human exposure to tsetse flies or mosquitoes, e.g., [151156], but they have not yet been applied on sand flies. Studies of antibodies’ reactivity to a single epitope could provide a better specificity and sensitivity when compared to whole SGH.…”

“…College Station, TX: StataCorp LP) and GraphPad Software V5. OD normalization and plate to plate variation was performed as described elsewhere (25). Briefly, antibody levels were expressed as the ΔOD value: ΔOD = ODx − ODb, where ODx represents the mean of individual OD in both antigen wells and ODb the mean of the blank wells.…”

“…ELISA conditions were standardized as described elsewhere (24,25). Also, DBS samples were prepared as by eluting half of a card circle into 300uL of elution buffer (PBS 1X, Tween 20 0.05%) and incubated overnight at 4°C.…”
