An Extracellular Matrix‐Mimicking Hydrogel for Full Thickness Wound Healing in Diabetic Mice

Abstract: An extracellular matrix-mimicking hydrogel is developed consisting of a hyaluronan-derived component with anti-inflammatory activity, and a gelatin-derived component offering adhesion sites for cell anchorage. The in situ-forming hyaluronan-gelatin (HA-GEL) hydrogel displays a sponge-like microporous morphology. Also, HA-GEL shows a rapid swelling pattern reaching maximum weight swelling ratio within 10 min, while at the equilibrium state, fully swollen hydrogels display an exceedingly high water content with … Show more

“…1) H&E staining assay. The samples were fixed by 4% paraformaldehyde, embedded using the paraffin, and sectioned for the hematoxylin-eosin (H&E) stain experiment ( Qin et al, 2018 ). The infiltration of inflammatory cells, induction of epidermal hyperplasia and formation of blood vessels were evaluated within the H&E-stained sections (5 μm) and quantified as described previously ( Yang et al, 2021a ) ( Gao et al, 2019 ).…”

“…Thiolated hyaluronic acid (HASH) was obtained following a pre-established method (Supporting Information Scheme S2 ) 24 . The obtained product (yield: 89.9%) was validated by 1 H NMR (400 MHz, D 2 O, δ ): 4.38 (bs, 2H), 3.0‒3.8 (bs, 5H), 2.54 (m, 2H), 2.30 (m, 2H), 1.87 (bs, 3H) (Supporting Information Fig.…”

“…[ 26,33 ] To introduce sulfhydryl (─SH) groups, reagents containing either free thiol or a disulfide bond are used to modify the carboxyl groups (Figure 2). [ 26,34 ] To introduce the double bond structure for further crosslinking, methacrylic anhydride is used to react with the methyl hydroxy group on the N‐acetyl‐D‐glucosamine (Figure 2). [ 35,36 ] To form a polyvalent electrophile, bromoacetic anhydride and 6‐hydroxy groups of the N‐acetylglucosamine residues are linked through amide reaction (Figure 2).…”

“…As 3D polymer networks, hydrogels display high water content, which are often used for localized and controlled drug delivery. [ 34 ] To construct the 3D structure, polysaccharides are chemically modified to introduce moieties that can form crosslinks between linear polysaccharide chains. In general, physical and chemical crosslinks have been widely explored in the literature to present hydrogel networks with different structures and functionalities.…”
