An essential role of NAD(P)H oxidase 2 in UVA-induced calcium oscillations in mast cells

Abstract: Solar UVA radiation (320-400 nm) is known to have immunomodulatory effects, but the detailed mechanisms involved are not fully elucidated. UVA irradiation has been shown to induce calcium oscillations in rat peritoneal mast cells due to NAD(P)H oxidase (NOX) activation, but the specific NOX isoforms have not been identified. In the present work effects of UVA irradiation were investigated in isolated rat peritoneal mast cells, in cultured rat mast cell line RBL-2H3, and in mouse bone marrow-derived mast cells … Show more

“…31 MC activation and proliferation are associated with enhanced reactive oxygen species (ROS) production mainly by using NADPH oxidase 2. 32,33 Furthermore, ROS play an important role in SM progression because they induce expression of the antiapoptotic protein DJ-1 in clonal MCs 34 ROS generation associated with MC proliferation could impair endotheliummediated dilation through several mechanisms, including (1) inhibition of vasodilator prostacyclin production, (2) uncoupling endothelial NO synthase, and (3) binding of NO with subsequent reduction of its bioavailability. 35 In patients with SM, univariate analysis shows that age, high serum tryptase levels, and flushing are associated with worse FMD, whereas MAP is directly related to FMD.…”

“…MCs, like keratinocytes, fibroblasts and several other cutaneous cells are also susceptible to low doses of narrowband (NB) UVB, making them targets of photons that lead to DNA damage and apoptosis . NAD(P)H oxidase 2 has been found important in UVA‐induced calcium oscillations and mediator production of mast cells . Whether or not MCs express photoreceptors, such as cryptochromes or opsins, remains to be investigated in detail.…”

“…Therefore, NOX2 and NOX2-associated signaling molecules may be good candidates for the targeted therapy of OS. NOX2 has been previously reported to be involved in a variety of physiological and pathological conditions, including prion disease (44)(45)(46)(47). NOX2 has also been implicated in cancer biology (48)(49)(50) as NOX2 was established to serve a pro-survival role in human leukemia and be involved to promote apoptosis in human glioma (48,50).…”
