An electrochemical immunosensing system on patterned electrodes for immunoglobulin E detection

Abstract: We present an electrochemical biosensing platform which enables immunoglobulin E (IgE) detection from eight independent biological samples. The performance of the electrochemical immunosensor was comparable to the conventional optical assay.

“…In this research, we introduced the electron transfer mediator, Fc, as a functionality in the APTES SAMs-modified ITO electrode. In previous studies [33][34][35], the Fc-modified SAMs were constructed on the Au electrode by a complicated method: for example, using a specially designed Fc-derivative, such as an Fc-tethered dendrimer or Fc-functionalized thiols. Therefore, less reproducible mixed SAMs using multiple thiols and additional immobilization steps using a crosslinker were required for the introduction of the Fc moiety in the electrochemical sensing layer.…”

“…The Nernst equation defines how the concentration of the species in solution near the working electrode changes over time [52]. Oh et al applied CV to an immunosensing platform for detecting IgE in multiple samples simultaneously [53]. A gold electrode surface was treated with a mixed self-assembled monolayer (SAM) containing 11-mercapto-1-undecanol (MUO), 12-mercaptododecanoic acid (MDA) and 11-(mercaptoundecyl)ferrocene (MUF).…”

“…Driven by this conviction, this work reports the first electroanalytical bioplatform reported so far for the single and simultaneous determination of OVA-specific IgE and IgG4. It is important to note that although some electrochemical biosensors for IgEs have been previously reported, 1,30-33 some of them require a long preparation time 32,33 or have limited sensitivity 31 and none of them dealt with the determination of specific IgE against food allergens. In addition, to our knowledge, no electrochemical bioplatform has been reported so far for the determination of IgG4 or targeting the simultaneous determination of different Igs related to allergic processes.…”
