Inbred strains are genetically stable across time and laboratories, allowing scientists to accumulate a record of phenotypes, including physiological characteristics and behaviors. To date, the C57/C58 family of inbred mouse strains has been identified as having the highest innate ethanol consumption, but some lineages have rarely or never been surveyed. Thus, the purpose of the present experiment was to measure ethanol preference and intake in 22 inbred mouse strains, some of which have never been tested for ethanol consumption. Male and female mice (A/J, BALB/cByJ, BTBR+T tf/tf , BUB/ BnJ, C57BL/6J, C57BLKS/J, C58/J, CZECH/Ei, DBA/2J, FVB/NJ, I/LnJ, LP/J, MA/MyJ, NOD/ LtJ, NON/LtJ, NZB/B1NJ, NZW/LacJ, PERA/Ei, RIIIS/J, SEA/GnJ, SM/J, and 129S1/SvlmJ) were individually housed and given unlimited access in a two-bottle choice procedure to one bottle containing tap water and a second containing increasing concentrations of ethanol (3%, 6%, 10%), 0.2% saccharin, and then increasing concentrations of ethanol (3%, 6%, 10%) plus 0.2% saccharin. Mice were given access to each novel solution for a total of 4 days, with a bottle side change every other day. Consistent with previous studies, C57BL/6J (B6) mice consumed an ethanol dose of > 10g/kg/day while DBA/2J (D2) mice consumed < 2g/kg/day. No strain voluntarily consumed greater doses of ethanol than B6 mice. While the C58 and C57BLKS strains showed high ethanol consumption levels that were comparable to B6 mice, the BUB and BTBR strains exhibited low ethanol intakes similar to D2 mice. The addition of 0.2% saccharin to the ethanol solutions significantly increased ethanol intake by most strains and altered the strain distribution pattern. Strong positive correlations (rs ≥ 0.83) were determined between consumption of the unsweetened versus sweetened ethanol solutions. Consumption of saccharin alone was significantly positively correlated with the sweetened ethanol solutions (rs = 0.62 -0.81), but the correlation with unsweetened ethanol solutions was considerably lower (rs = 0.37 -0.45). These results add new strains to the strain mean database that will facilitate the identification of genetic relationships between voluntary ethanol consumption, saccharin preference, and other phenotypes.

Infection of inbred mouse strains with Citrobacter rodentium represents an ideal model to reveal the genetic factors controlling host resistance to noninvasive enteric bacterial pathogens. We have chosen a positional cloning approach to identify putative gene(s) that control the known difference in survival between resistant C57BL/6J and susceptible C3H/HeJ and C3H/HeOuJ mice. Our work has identified one major locus within proximal chromosome 15 that is responsible for the marked susceptibility of both C3H strains, and we formally exclude Tlr4 from control of survival to this pathogen. We have named this new host resistance locus Cri1 (Citrobacter rodentium infection 1). The Cri1 genetic interval currently spans B16 Mb and it confers survival to the infection in a recessive manner. Transfer of the Cri1 locus from the surviving B6 mice into a congenic mouse with a C3Ou genetic background confirms its overall chromosomal localization and its highly significant effect on host survival. The C3Ou.B6-Cri1 mice thus produced have also enabled us to dissociate the control of mouse survival from the control of bacterial load early in the infection as well as from control of colonic hyperplasia.

We measured voluntary calcium intake, blood calcium, and bone mineral content of male and female mice from 40 inbred strains. Calcium intakes were assessed using 48-h two-bottle tests with a choice between water and one of the following: water, 7.5, 25, and 75 mM CaCl 2 , then 7.5, 25, and 75 mM calcium lactate (CaLa). Intakes were affected by strain, sex, anion, and concentration. In 11 strains females consumed more calcium than did males and in the remaining 29 strains there were no sex differences. Nine strains drank more CaLa than CaCl 2 whereas only one strain (JF1/Ms) drank more CaCl 2 than CaLa. Some strains had consistently high calcium intakes and preferred all calcium solutions relative to water (e.g., PWK/PhJ, BTBR T + tf/J, JF1/Ms). Others had consistently low calcium intakes and avoided all calcium solutions relative to water (e.g., KK/H1J, C57BL/10J, CE/ J, C58/J). After behavioral tests, blood was sampled and assayed for pH, ionized calcium concentration, and plasma total calcium concentration. Bone mineral density and content were assessed by DEXA. There were no significant correlations between any of these physiological measures and calcium intake. However, strains of mice that had the highest calcium intakes generally fell at the extremes of the physiological distributions. We conclude that the avidity for calcium is determined by different genetic architecture and thus different physiological mechanisms in different strains. Blood ionized calcium may regulate calcium intake [30,51,54], perhaps by acting on calcium receptors in the brain [e.g., [32]]. Circulating levels of parathyroid hormone, calcitonin, and 1,25-dihydroxyvitamin D influence calcium intake [e.g., [12,30,43,51,61]] but it is unclear whether these effects are due to the direct action of the hormones on the brain or to their indirect actions on blood calcium levels. The most-studied control of calcium intake involves orosensory acceptance. Calcium deficiency lowers the behavioral and electrophysiological detection thresholds for calcium [25]. It also increases the avidity for concentrations of calcium that are disliked in the replete state: calcium deficiency increases calcium intakes in preference tests [14,25], the rate of shamingestion of calcium [34], the frequency of hedonically positive facial expressions during calcium consumption [33], and the response to oral calcium of sweet-best units in the nucleus of the solitary tract [35].
