Background . In Laos, the malaria burden remains high despite a significant reduction of cases during the last decade. In the context of the disease elimination by 2030, a nationwide entomological survey was conducted to better understand the distribution, abundance and behavior of major malaria vectors ( Anopheles spp.) in the country. Methods . Mosquito collections were implemented in ten villages from ten provinces during the rainy and dry seasons of 2014 and 2015 by using human landing catch (HLC) and cow bait collection (CBC) methods. After morphological identification in the field, molecular identification of the sibling species of Anopheles mosquitoes from the Funestus, Leucosphyrus, and Maculatus groups were determined using PCR specific alleles. A screening of Plasmodium falciparum and P. vivax infections in the vectors was carried out by quantitative PCR assays. Results. A total of 14,146 adult mosquitoes representing 25 different Anopheles species were collected and morphologically identified. Molecular identification revealed the presence of 12 sibling species within the main primary vector groups including An. maculatus s.s., An. rampae , An. sawadwongporni , An. pseudowillmori , An. dravidicus , An. minimus s.s., An. aconitus , An. pampanai , An. harrisoni , An. dirus s.s., An. baimaii , An. nemophilous . Anopheles maculatus and An. minimus were predominant during both the dry and rainy seasons, but showed highly zoophilic preferences (Zoophilic index of 98% and 95%, respectively). Overall, 22% of the total malaria vectors were collected between 10:00 PM and 5:00 AM indoors when people are sleeping. Twenty-seven percent of primary and secondary vectors were collected outdoors before 10:00 PM or after 5:00 AM, times when people are usually awake and outdoors. Only two specimens were positive for P. falciparum , one An. aconitus from Phongsaly and one An. minimus from Vientiane Province Conclusions. The results indicate that people living in rural areas in Laos are constantly exposed to malaria vectors throughout the year and specifically outdoors. The use of LLINs/IRS remains important but innovative tools and new strategies are needed to address locally, the early and outdoor malaria transmission. Lack of expertise in general entomological methods may further exacerbate the situation.

Mosquitoes are major vectors of many infectious pathogens or parasites. Understanding cryptic species and the speciation of disease vectors has important implications for vector management, evolution and hostpathogen and/or host-parasite interactions. Currently, mosquito cryptic species have been reported in many studies, most of which focus on the reproductive isolation of cryptic species and mainly on Anopheles gambiae sensu lato complex. Emerging species within the primary malaria vector Anopheles gambiae show different ecological preferences and significant prezygotic reproductive isolation, while Aedes mariae and Aedes zammitii show postmating reproductive isolation. However, data reporting the reproductive isolation in Culex and Aedes albopictus mosquito cryptic species is absent. The lack of systematic studies leaves many questions open, such as whether cryptic species are more common in particular habitats, latitudes or taxonomic groups; what mosquito cryptic species evolutionary processes bring about reproductive isolation in the absence of morphological differentiation? How does Wolbachia infection affect in mosquitoes' reproductive isolation? In this review, we provide a summary of recent advances in the discovery and identification of sibling or cryptic species within mosquito genera.

Understanding the processes of reproductive behaviour in mosquitoes is crucial for improving mating competitiveness and mating specificity for sterile insect release programmes. The Culex pipiens (Linneaus) (Diptera: Culicidae) forms pipiens and molestus (Forskål), two biotypes of the Cx. pipiens complex, are vectors for West Nile virus, St Louis encephalitis virus and lymphatic filariases. Hybridization of these biotypes is known to occur in nature, although form pipiens mates above ground in large spaces (eurygamy) and form molestus preferentially mates in small spaces (stenogamy) such as sewage tunnels. Hybridization may allow gene flow of biotype-specific characteristics that are crucial in the disease transmission cycle. The present study examined and compared mating behaviours, insemination rates, fecundity and fertility in parental and F1 hybrids between Cx. pipiens f. pipiens and Cx. pipiens f. molestus in conditions of stenogamy. Unique mating behaviour sequences were identified in Cx. pipiens f. molestus, including tapping, mounting, co-flying and copulation. Despite the considerably high insemination rates in hybrid crosses, fertility and fecundity rates were varied. This observation could suggest reproductive isolation in the hybrid zone. The study also documents a failure of heterospecific males to produce fertile eggs in Cx. pipiens f. pipiens females, which may be attributable to gametic incompatibilities and may represent an additional barrier to gene exchange.
