An astrovirus frameshift signal induces ribosomal frameshifting in vitro

Abstract: Expression of the astrovirus RNA-dependent RNA polymerase has been hypothesized to be regulated by (-1) ribosomal frameshifting. Sequence analysis of the 70 nucleotide region between open reading frames 1a and 1b indicates the presence of a shifty heptamer consensus sequence and downstream sequences that may be needed for ribosomal frameshifting. We constructed four astrovirus cassettes that spanned this region and inserted each into the rhesus rotavirus VP4 gene. The constructs were expressed in an in vitro s… Show more

“…The 76-kDa band may be explained by the occurrence of a frameshift after aa 903 of nsP1a that led to the translation of the 122 ORF1b-derived amino acids followed by 11 vector-encoded amino acids (expected molecular mass of the fusion protein, 72 kDa). The prominence of the 76-kDa band suggests that during translation of His-394-nsP1a, frameshifting occurs at a frequency higher than the 5 to 7% reported previously (13). This may be due to an altered conformation of the nascent His-394-nsP1a during translation compared to that of the full-length nsP1a.…”

“…The translation product of ORF1a is designated nonstructural protein 1a (nsP1a). Expression of ORF1b, which is in a Ϫ1 position relative to ORF1a, depends on the occurrence of a Ϫ1 ribosomal frameshift during the translation of ORF1a (5 to 7% frequency in a cell-free, uncoupled transcription-translation system [13]), leading to an nsP1a/1b fusion protein (13,14). ORF2 encodes the viral capsid protein and is expressed from a subgenomic RNA colinear with the 3Ј one-third of the genomic RNA (12,17).…”

“…The ribosomal frameshift sequence linking ORF1a and 1b is a feature typical of astroviruses (Lewis & Matsui, 1995, 1996, 1997Marczinke et al, 1994), as are the amino acid motifs for a serine protease in ORF1a and the RdRp of the picornavirus supergroup lacking a helicase motif in ORF1b. Comparisons of the deduced amino acid sequences from ORF1a, 1b and 2 of MiAstV with the corresponding ORFs of its closest relative, OAstV, revealed identities of 38, 63 and 37 %, respectively.…”

“…ORF1a encodes a polypeptide of 920 to 935 amino acids (aa) containing several conserved motifs, including a serine protease (11,44). A highly conserved heptanucleotide motif and downstream hairpin structure at the ORF1a-ORF1b junction generates a Ϫ1 frameshift, leading to the translation of an ORF1a-ORF1b polypeptide which is later cleaved into polypeptides corresponding to ORF1a and ORF1b (11,29,41,44). ORF1b encodes a polypeptide of approximately 515 to 528 aa, containing the RNA-dependent RNA polymerase (RdRP) (40,44).…”
