An Assessment of New Sperm Tests for Male Infertility

Abstract: The routine semen analysis, although used for more than 50 years, fails to accurately distinguish between fertile and infertile men. As a consequence, many tests of sperm function (TSF) have been developed. This review discusses both older and newer diagnostic TSF. It outlines the principles underlying each assay and reviews aggregate clinical data to determine its current relevance and utility. It concludes that the relevance of many older TSF is questionable, with the wide acceptance of intracytoplasmic sper… Show more

“…Infertility is reported to be on the increase over the past decade due to the number of couples seeking consultation for infertility problems with about 50% attributed to male factors [2,3]. Infertility among males could be basically diagnosed through semen analysis which involves the evaluation of sperm count, sperm motility, and sperm morphology [4,5,6,7]. Quite a number of factors have been reported to be actively associated with infertility.…”

“…At present, the diagnosis of male infertility has mainly been based on traditional semen parameters, including seminal vol-ume, pH, sperm concentration, motility, and morphology, as recommended by the WHO (6 ). Routine semen analysis, although in use for Ͼ50 years, has failed to distinguish accurately between fertile and infertile men (7)(8)(9). In addition, because seminal plasma contains many biochemical molecules secreted from the specific organs or compartments of the male genital system, changes in some factors, including acid phosphatase, ␣-glucosidase, and others, have also been used as diagnostic indicators for evaluating the male reproductive system.…”

“…Foram identificadas quatro principais causas de quebras na estrutura do dna: 1) apoptose abortiva durante mitose inicial, 2) rupturas dos filamentos de dna não resolvidas e criadas durante a espermatogênese para aliviar as tensões da torção associadas à remodelação da cromatina, 3) maturação espermática anormal causada por distúrbios na protaminação e 4) estresse oxidativo derivado da liberação de radicais livres provenientes do metabolismo celular [70,71].…”
