An ALP-activatable and mitochondria-targeted probe for prostate cancer-specific bimodal imaging and aggregation-enhanced photothermal therapy

Abstract: We developed a mitochondria-targeted near-infrared activatable fluorescent/photoacoustic (NIR FL/PA) probe for the selective detection of prostate cancer-derived ALP and aggregation-enhanced photothermal therapy.

“…The photothermal conversion capability of small molecule PTAs was regulated by the electrical changes initiated from the regulation of specic substances in tumor cells. 113 An aggregation-enhanced PTT probe (termed as ETP) was developed by Yao et al for mitochondria-targeted imaging and therapy, 98 which was activated by alkaline phosphatase (ALP) (Fig. 6a right).…”

“…In addition to the above‐introduced ALP‐activatable probes employed for imaging, theranostic systems triggerable by ALP have also been reported. [ 90 ] The enzyme‐activatable theranostic probe ETP is composed of a NIR emitter, namely the hemicyanine dye, a mitochondria‐targeting hydrophobic triphenylphosphonium group, and a phosphate ester group. In this probe, the hemicyanine contributes to the NIR FL/PA imaging and phototherapy, and the phosphate ester unit works not only as the ALP substrate but also as a quencher to the FL/PA signals of the hemicyanine in its original form.…”

“…[87] In view of its overexpression in tumors and its importance in early diagnosis and therapy of cancers, such as metastatic prostate cancer, several ALP-activatable probes have been explored for the specific imaging or theranostics of cancer. [88][89][90][91][92][93] 24 and 25 are displayed in Figure 12 as representatives of the developed ALPtriggerable probes.…”

“…In another design, Zhang and co‐workers reported a prostate cancer imaging platform using an ALP‐activatable and mitochondria‐targeted probe. [ 78 ] The FL and PA intensity of the probe (ETP), composed of a NIR FL dye, a lipophilic triphenylphosphium cation that targets mitochondria, and a phosphate ester group, was drastically increased after incubating with ALP. The enhancement was attributed to the removal of the phosphate group that quenched FL/PA signal and resulted in situ NPs self‐assembly.…”
