Gills of fish are involved in respiration, excretion and osmoregulation. Due to numerous interactions between these processes, branchial diseases have serious implications on fish health. Here, "koi sleepy disease" (KSD), caused by carp edema virus (CEV) infection was used to study physiological, immunological and metabolic consequences of a gill disease in fish. A metabolome analysis shows that the moderately hypoxic-tolerant carp can compensate the respiratory compromise related to this infection by various adaptations in their metabolism. Instead, the disease is accompanied by a massive disturbance of the osmotic balance with hyponatremia as low as 71.65 mmol L −1 , and an accumulation of ammonia in circulatory blood causing a hyperammonemia as high as 1123.24 µmol L −1 . At water conditions with increased ambient salt, the hydro-mineral balance and the ammonia excretion were restored. Importantly, both hyponatremia and hyperammonemia in KSD-affected carp can be linked to an immunosuppression leading to a four-fold drop in the number of white blood cells, and significant downregulation of cd4, tcr a2 and igm expression in gills, which can be evaded by increasing the ion concentration in water. This shows that the complex host-pathogen interactions within the gills can have immunosuppressive consequences, which have not previously been addressed in fish. Furthermore, it makes the CEV infection of carp a powerful model for studying interdependent pathological and immunological effects of a branchial disease in fish.

Nine experimental diets were formulated to study the effects of fishmeal replacement on Opsariichthys bidens. Weight gain rate, protein efficiency rate and apparent digestibility coefficient of fish in fishmeal (FM), maggot meal (MM) and soybean meal (SM) groups were significantly the highest. There was no significant difference in intestinal amylase activity between FM and SM groups. Principal component analysis showed that SM group and wild fish (WF) individual had similar essential amino acid compositions in muscle. The lowest value of serum malondialdehyde and the highest value of total antioxidant capacity were found in SM group, but fish in MM group did the opposite. Fish in MM group had the higher whole-body lipid, serum triglycerides, glucose, complement C3, C4 and immunoglobulin contents and lysozyme activity. After ammonia exposure, cumulative mortality of fish in FM and SM groups was significantly the lowest. Liver carbamyl phosphate synthetase activity of fish in FM group was significantly the highest, but the highest values of glutamine synthetase and glutamate dehydrogenase were found in SM group. This study suggests that fishmeal replacement with soybean meal is acceptable, and muscle amino acid compositions of fish in SM group were more similar to wild individual.

In this study, nine experimental diets were formulated to contain three graded levels of protein (40, 45 and 50% of dry diet) and three graded levels of cellulose (0, 5 and 10% of dry diet). These diets were then fed in triplicate to yellow catfish for 90 days. We found that fish fed diets containing 45% and 50% protein had more weight gain than did fish fed a 40% protein diet, while fish fed diets supplemented with 5% cellulose achieved the greatest weight gain and the lower feed conversion ratio. We found that as dietary cellulose and protein levels were increased, the activities of antioxidant enzymes (superoxide dismutase, catalase and glutathione peroxidase), immune response parameters (alkaline phosphatase, acid phosphatase, phagocytic index, respiratory burst, complement 3 and 4) and the mRNA level expression of copper/zinc superoxide dismutase, catalase, glutathione peroxidase and glutathione reductase all rose. In contrast, rising cellulose levels were associated with decreased malondialdehyde contents and increasing tumour necrosis factor, interleukin 1 and interleukin 8 mRNA expression. Overall, our results indicate that dietary protein supplementation can enhance yellow catfish immune functionality and that while dietary protein impacts on the immune response are influenced by dietary cellulose levels, increased dietary cellulose was associated with inflammatory gene upregulation.
