Amino acid profiling in plant cell cultures: An inter‐laboratory comparison of CE‐MS and GC‐MS

Abstract: A CE-MS method for metabolic profiling of amino acids was developed and used in an integrated functional genomics project to study the response of Medicago truncatula liquid suspension cell cultures to stress. This project required the analysis of more than 500 root cell culture extracts. The CE-MS method profiled 20 biologically important amino acids. The CE-MS method required no sample derivatization prior to injection and used minimal sample preparation. The method is described in terms of CE and MS operati… Show more

“…3. The CE-MS operation has also been found to be less expensive than GC-MS since one cheap fused-silica capillary can be used in over 1500 injections [122]. Lee et al [123] used CE-MS in conjugation with computer simulation to analyze low-abundance metabolites from complex samples.…”

“…Using DNA microarray technology, 2-D differential gel electrophoresis, and CE-TOF MS, this group showed the effect of perturbation at all omics levels. Williams et al [122] reported on the analysis of important amino acid profiles in different plant cells. In that report the authors concluded that CE-MS analysis could be a valid technique for profiling metabolites, and that it had advantages over GC-MS, a common analytical technique in this area, because it did not require sample derivatization, thereby making sample preparation simpler.…”

“…Although the application of CE-MS to plant metabolite profiling is relatively limited (Sato et al, 2004;Takahashi et al, 2006;Williams et al, 2007), CE-MS will be used in a broader field of application due to its applicability to the polar metabolites and ease of sample preparation.…”

“…Nevertheless, both techniques yielded the same biological conclusions on environmentinduced changes in plant amino-acid levels. TMS derivatives with GC-MS analysis of amino acids was further compared to capillary electrophoresis coupled to MS, demonstrating that both techniques obtain similar results [34].…”
