Preparation and chemical investigation of Ficus carica extract The leaves of Ficus carica were collected from Giza governorate, Egypt, in Sep. 2018, and identified by ABSTRACT Objective: The present work investigated the protective effect of Ficus carica (common fig) leaves methanol extract against genotoxicity and testicular damage of cisplatin (CP) and identified some of its active ingredients. Methods: Seven main groups were investigated as follows: I. control negative, II. Control plant (600 mg/kg fig, orally), III, IV. Control positive (treated i.p with 10 and 15 mg/kg CP), V-VII. groups treated with fig (200, 400 and 600 mg/ kg) + Cisplatin (15 mg/kg). Results: Ficus carica alleviated the destructive effects of CP in the testis, liver and bone marrow due to the presence of high amount of flavonoids and phenolic compounds. Also it has a normal effect in the tested parameters as compared with the control negative. Chromatographic investigation resulted in the identification of 6 compounds: Catechin, Luteolin-8-C-β-D glucopyranoside, Quercetin, Quercetin-3-O-β-d-glucopyranoside, Chlorogenic acid and Kaempferol-3-O-β-D-glucopyranoside. In bone marrow cisplatin induced significant percentage of chromosome abnormalities, micronuclei in polychromatic erythrocytes and toxicity to cells. On the contrary the two tested doses of cisplatin had a normal effect on spermatocyte chromosomes (germ cells). The dose 15 mg/kg induced an overexpression of the liver genes NF-kB and iNOS as indicated by real-time PCR. Different forms of histopathological alterations and instigation of the expression of TNF-α gene in the testis were detected after CP treatment. Conclusion: Ficus carica is a promising candidate rich in many bioactive constituents and can be used in combination with chemotherapeutic drugs to alleviate their destructive effects.

“…In another cisplatininduced testicular toxicity study it was found that only cisplatin treatment resulted in a significant decrease in daily sperm production, head length and % DNA in head, reduction of epithelial cells height, tubular diameter, number of spermatogonia, spermatocytes and spermatids, the intratesticular testosterone concentrations, increase in the thiobarbituric acid reactive substances (TBARS) and oxidative stress in testicular tissues. Rutin treatment to cisplatin-treated rats resulted in reversing cisplatin's effects on DNA damage, sperm count, histological and biochemical parameters, thus suggesting the shielding effects of rutin against cisplatin-induced testicular toxicity (Jahan et al, 2018). The effects of rutin on diabetic-induced impairments in sexual behaviour, spermatogenesis and oxidative testicular damage have also been reported.…”

Nature is an attractive source of new therapeutic agents and a variety of phytochemicals have proved to be very valuable in the prevention and treatment of various diseases. Many of the important drugs isolated from plants during the past 50 years have revolutionized modern medicinal practice (Dar et al., 2017). The use of herbal medicines and phytonutrients or nutraceuticals continues to expand rapidly across the world with many people now resorting to these products for the treatment of various health challenges in different national healthcare settings (WHO, 2004). It is estimated that up to 80% of the world's population living in the developing countries rely on herbal medicinal products as a primary source of healthcare and traditional medical practice as an integral part of the culture in their communities (Ekor, 2013). The potential testicular protective effect of nimesulide (NSE) was studied in cisplatin (CSP)-challenged rats. NSE therapy (10 mg/kg/day, p.o.) was applied for 15 days, and a single dose of CSP (7 mg/kg, i.p.) was administered on the 10th day. CSP significantly decreased the levels of serum testosterone, testicular reduced glutathione, and superoxide dismutase. CSP also significantly increased testicular malondialdehyde, nitric oxide, tumor necrosis factor-α, interleukin-1β, cyclooxygenase-2 (COX-2), prostaglandin E2, nuclear factor-κB p65, Bax, and caspase-3. NSE significantly ameliorated all the biochemical changes observed in CSP-challenged rats. Moreover, NSE significantly reduced the histopathological injury, and the expressions of phosphorylated c-Jun N-terminal kinase (p-JNK) and p38 mitogen-activated protein kinases (MAPKs) in testes of rats received CSP. It was concluded that NSE significantly blocked the CSP-induced acute testicular injury in rats through inhibition of MAPK/COX-2 signaling pathway, and by combating oxidative stress, inflammation, and apoptosis.
