Alternative to FBS in animal cell culture - An overview and future perspective

Abstract: Fetal bovine serum (FBS) is a widely used growth supplement in the in vitro culturing of animal and human cells, tissues and organs, notably due to the occurrence of abundant micro-and macronutrients, along with growth factors. Over the years, increasing demand, high price, batch-to-batch variability in quality and composition, increasing ethical concerns lead to the search for an alternative to FBS. Several approaches have been suggested and employed in the past, but none is implemented as widely as FBS, and … Show more

“…Figure 5C shows the increase in cell number and metabolism over time in cells seeded with culture media 2% FBS. On the contrary, the cells seeded with culture media without FBS show a stationary behaviour in their metabolism and viability over the 5 h. These results are in agreement with the fact that FBS supplementation improve cell viability and metabolism ( Subbiahanadar Chelladurai et al, 2021 ). Furthermore, the plasmonic biosensor can accurately detect the cell adhesion and confluency by monitoring the free gold surface available.…”

“…Fetal bovine serum (FBS) is often used in research in a broad range of cell cultures as a source of nutrients and growth factors. However, it has several issues: firstly, FBS does not simulate the human microenvironment, which limits its translational application ( 131 ); secondly, it involves the risk of transmitting bovine spongiform encephalopathy and some viral infections; thirdly, it can promote the development of unwanted immunological reactions; and, finally, variations between the brands and batches of sera can affect the reproducibility of the experiments ( 131 , 133 , 134 ). Human serum (HS) does not contain any xenogeneic components and supplements the medium with trophogens and additional stimuli that favor cell growth and survival.…”
