Altered expression of miRNA-155 and miRNA-21 and higher numbers of T cells in the peripheral blood of early diagnosed lung cancer patients

Abstract: Background: MicroRNAs (miRNAs) play a significant role in shaping immune response. The expression profile of miRNAs, however, has been found to be affected by the presence of tumor, but whether the incidence of non-small cell lung cancer (NSCLC) alters the expression profile of miRNAs has not been studied. Objective: The current study aims to investigate the expression of miRNAs in the peripheral blood mononuclear cells (PBMCs) from patients with NSCLC as well as to analyze the numbers of helper CD4 + and cyto… Show more

“…The miR-21-3p (1.65-fold change) and miR-21-5p (1.82-fold change) are both up-regulated in PBMCs of NSCLC patients, corroborating with miR-21 expression determined in plasma [ 39 , 40 , 41 , 42 , 43 ]. Our results correlate well with those attained in previous studies showing over-expression of miR-21 in NSCLC PBMCs as a possible biomarker [ 25 , 44 ].…”

“…Malignant NSCLC tissue shows dysregulated miRNA profiles in comparison with control counterparts via molecular analysis [ 21 , 22 ]. Plasma, serum and sputum constitute important biological samples for NSCLC-miRNA screening, concerning a less invasive sample collection [ 23 , 24 , 25 ]. Peripheral blood mononuclear cells (PBMCs) are arising as a potential source of significant biomarkers, including miRNAs [ 26 ].…”

“…In contrast to such an undetectable phenomenon, the further execution of the NIR photocontrolled sensing gives rise to noteworthy fluorescence brightness (originating from the Cy5 signal recovery) in both live cancer (A549 and MCF-7) and normal (L02) cells (top panel in Figure A), implying that the whole sensing flow works well in cellular media. In addition, on account of the overexpression essence of miRNA-155 in lung cancer, , it is not difficult to observe that the strongest Cy5 signal belongs to the A549 group, and a progressive downward trend is assigned in turn to the MCF-7 and L02 groups. Quantifying the relevant fluorescence images using Image-J software determination (the rightmost panel in Figure A) results in a discovery that the average fluorescence intensities (120 cells are analyzed for each group) of the NIR light-treated cases are in the following order ∼4.61 times (A549), ∼3.17 times (MCF-7), and ∼1.89 times (L02) more than the corresponding unirradiated cases (Figure B).…”

“…In accordance with the as-mentioned inspirations, we aim to construct a fluorescent nanosensor by combining a NIR photocontrolled sensing notion with a six-branched DNA nanowheel-induced nucleic acid confinement effect. To examine the bioimaging capability in live biosystems, a short noncoding microRNA sequence (miRNA-155) owning the overexpression tendency in lung cancer , is selected as a model low-abundance analyte. In our establishment flow (Scheme A), a satellite-like nanoframe (termed SLN) is ultimately fabricated by employing a covalent 1-ethyl-3-(3-dimethylamino-propyl)carbodiimide hydrochloride (EDC)-activated bio-coupling between a photocontrolled DNA composite precursor (termed as PCDCP, labeled with an amino group at its 5′ end) and a polyacrylic acid (PAA) polymer-functionalized upconversion nanoparticle (termed as UCNP-PAA).…”

“…It promotes cell growth, metastasis, and chemotherapy or radiotherapyresistance of NSCLC cells by targeting tumor suppressor phosphatase and tensin homolog (PTEN) (Liu et al, 2013). Also, miRNA-21 is greatly expressed in the serum of NSCLC patients and low miRNA-21 expression can decrease malignant cell proliferation, migration, and invasion (Zhu et al, 2008;Yang Y et al, 2015;Zhu et al, 2017;Salem et al, 2020). miRNA-98, a let-7 family member is thought to be associated with many tumors.…”
