Altered expression of Butyrophilin (BTN) and BTN‐like (BTNL) genes in intestinal inflammation and colon cancer

Abstract: Several Butyrophilin (BTN) and Btn‐like (BTNL) molecules control T lymphocyte responses, and are genetically associated with inflammatory disorders and cancer. In this study, we present a comprehensive expression analysis of human and murine BTN and BTNL genes in conditions associated with intestinal inflammation and cancer. Using real‐time PCR, expression of human BTN and BTNL genes was analyzed in samples from patients with ulcerative colitis, irritable bowel syndrome, and colon tumors. Expression of murine … Show more

“…For real-time PCR analysis, 1.5 μg of total RNA was reverse transcribed into cDNA using a high-capacity RNA-to-cDNA Kit (Applied Biosystems) according to the manufacturer’s instructions. Expression levels of three mucin genes ( MUC2 , MUC4 , and MUC5AC ; TaqMan assay IDs Hs.PT.56a.26485553, Hs.PT.56a.5039491 and Hs.PT.56a.25473826, respectively), relative to the reference gene hypoxanthine phosphoribosyltransferase [ 42 , 43 ] ( HPRT1 ; Hs.PT.39a.22214821, NM_000194.1), were determined using TaqMan probes on the Rotor-Gene 6000 real-time thermal cycler (Corbett Life Science, Concord, Australia). All PCRs (no template controls, untreated, and treated samples) were prepared as triplicate 10 μL reactions as described previously [ 44 ].…”

“…[118] Gene expression analysis of biopsies from ulcerative colitis patients showed increased BTN3 and decreased BTNL8 expression. [119] Based on the function of these genes, these changes in expression correlate with a suppressive effect on T cell activation;[119] however, the effect that this has on γδ IEL activation remains to be determined.…”

“…It has been localized to Peyer's patches of the small intestine as well as epithelial and dendritic cells within gastrointestinal tissue. Altered expression of BTNL2 is associated with inflammatory bowel disease [14]. Mutations in this gene are further associated with other autoimmune conditions including sarcoidosis, rheumatoid arthritis, vitiligo, and psoriasis [15][16][17][18].…”
