Alterations of lipid composition in Friend leukemia cell tumors in mice treated with tumor necrosis factor‐α

Abstract: Lipid analyses were carried out on transplantable murine Friend leukemia cell tumors, 6 h after intratumoral administration of tumor necrosis factor-a (TNF). The levels of the major phospholipid classes were uniformly decreased to about 70% of control values; free fatty acids were increased to about 1705%; diacylglycerol was decreased to about 50% and triacylglycerol, the main lipid component, was not significantly altered. These results analysed in the light of concomitant alterations in the levels of phospho… Show more

“…Similar results have been reported by various authors (Iguchi et al, 1989;Kumar et al, 1991;Lin et al, 1992). Bravo et al(1990) found increased unsaturated FFA concentrations in transplantable Friend leukemic cell tumors, and someone believed that they represented a metabolic and energy substrate for the tumor itself (Spector, 1967). Morris hepatoma 7777 cells freshly isolated from highly malignant tumors grown in the hindlimb of buffalo rats actively convert ketone bodies to cholesterol and fatty acids.…”

“…Standards of phospholipids (phosphatidylcholine (PC); phosphatidylethanolamine (PE)), sphingomyelin (SM), triglyceride (TG), and cholesterol (Chol) were purchased from Sigma Aldrich Co. (St. Louis, Missouri, USA). Analysis of the fatty acid methyl esters of the single phospholipid classes was performed as already described with minor modifications [28]. Briefly, the fatty acid methyl esters were prepared by acid catalyzed trans-esterification [29] using heptadecanoic acid as internal standard, extracted in hexane and evaporated under nitrogen gas.…”
