Alloreactivity

Abstract: The alloimmune response between individuals genetically disparate for antigens encoded within the major histocompatibility complex (MHC) remains a substantial barrier to transplantation of solid organs, tissues, and hematopoietic stem cells. Alloreactivity has been an immunological paradox because of its apparent contradiction to the requirement of MHC restriction for the induction of normal T lymphocyte mediated immune responses. Through crystallographic analyses and experimental systems utilizing murine CD8(… Show more

“…Alloreactivity assay is relatively easy to perform and is commonly used to score APC function of cultured-expanded B cells. However, since the mechanism of alloreactivity is not entirely understood [49], APC function of cultured B cells should be further confirmed by antigen presentation to autologous T cells. Meanwhile, appropriate (positive and negative) controls should always be included in testing APC function and T cell responses to facilitate accurate interpretation.…”

“…1,2 In 1960s, the discovery of highly polymorphic genes tightly clustered (loci) explained how the T cells elicit a specific Immune response against to foreign antigens (alloreactivity) in specific cells. [3][4][5] The genes mentioned previously are termed as the Major Histocompatibility Complex (MHC) or Human Leukocyte Antigens (HLA) in Homo Sapiens. The molecule coded by MHC behaves as a platform for peptides processed the cells (antigenic presentation); thus, this allows the training of T lymphocytes for their selective recognition.…”
