Akt isoform-dependent regulation of ATP-Binding cassette A1 expression by apolipoprotein E

Abstract: We previously reported that apolipoprotein E (apoE) upregulates ATP-binding cassette transporter A1 (ABCA1) transcription through phosphatidylinositol 3-kinase (PI3K). Here we demonstrate that treatment of murine macrophages with human apoE3 enhanced Akt phosphorylation, and upregulated ABCA1 protein and mRNA expression. Inhibition of PI3K weakened apoE3-induced Akt phosphorylation, and ABCA1 protein and mRNA increase. In contrast, inhibition of Akt only diminished apoE-induced ABCA1 protein but not the mRNA l… Show more

“…Akt increased the expression of ABCA1 by delaying its degradation, but does not alter the mRNA levels (41). The protective effect of the ABCA1 protein is mediated by Akt2, possibly by stabilizing ABCA1 resistance to calpain-mediated proteolysis.…”

“…This reduction in intracellular cholesterol enhances IL-4 receptor (IL-4R)/STAT6/PI3K signaling in vitro and in vivo, promoting the expression of the typical M2 marker arginase-1 while inhibiting NOS2 and proinflammatory IL-12 expression, thereby promoting tumor progression [ 309 , 310 ]. This cholesterol efflux is dependent on PI3K and Akt activation, which can upregulate ABCA1 expression in macrophages [ 309 , 311 ]. Moreover, cholesterol efflux leads to decreases in lipid rafts in macrophages, which act as signaling platforms for various receptors, including IFNγR, decreasing IFNγ-dependent expression of the proinflammatory genes IL-12 and iNOS [ 309 ].…”

“…S100B encodes a calcium-binding protein involved in neurite extension and axonal proliferation, ultimately being involved in synaptic plasticity and learning. Phosphorylation of Akt is enhanced by APOE (specifically, APOE3) ( Okoro et al, 2016 ), whereas APOE interacts with S100B during astrocytic activation/inhibition ( Mori et al, 2005 ). APOE encodes a component of the Reelin signaling pathway, which has been involved in verbal memory deficits in SZ ( Verbrugghe et al, 2012 ; Li et al, 2015 ).…”
