Age-Dependent Paraoxonase 1 (PON1) Activity and LDL Oxidation in Wistar Rats during Their Entire Lifespan

Abstract: Paraoxonase 1 (PON1) is an HDL bound enzyme which plays a key role in the protection of LDL and HDL from oxidation by hydrolyzing activated phospholipids and lipid peroxide products. Oxidative stress plays a crucial role in the development of atherosclerosis by oxidation of LDL. This study was conducted to determine age-dependent changes in plasma PON1 arylesterase activity and LDL oxidation in rats during their entire lifespan. 48 Wistar strain rats were grouped in six different age groups (1, 4, 8, 12, 18, a… Show more

“…Additionally, analysis of PON-1 concentrations/activities with respect to age and BMI, as factors that can affect PON-1 synthesis, showed an inverse relation of these parameters to age in the group of AP patients. It remains in accordance with the results of the study conducted by Kumar et al, in which a strong relation of age and PON-1 activity was shown 41. It was noted that age was strongly related to the increase of blood level of lipid peroxidation markers, which could impair PON‑1 function as antioxidant or its synthesis dependent on lipoprotein metabolism 41.…”

“…Lower levels of PON1 in C57BL/6J mice have been reported to result in lower levels of lipid metabolism intermediates, in comparison to suggesting a decrease in lipolysis and decreased fatty acid oxidation . A few reports show the role of polyphenols to stimulate PON1 . The maintenance of the enzyme‐SH group justifies the importance of plasma redox status as an important factor in the modulation of PON1 activity .…”

“…43 A few reports show the role of polyphenols to stimulate PON1. 44 The maintenance of the enzyme-SH group justifies the importance of plasma redox status as an important factor in the modulation of PON1 activity. 45 Our observation of hesperidin supplementation reversing the effect of hyperlipidaemic stress in atherogenic supplemented rats highlights the antioxidant role of hesperidin in protecting the -SH group of PON 1.…”

“…did not observe such a reduction in children [ 9 , 10 ]. This may be due to the decrease of enzyme activity with age [ 11 ]. Our results are consistent with those of Aslan et al .…”
