Age-dependent frequency of unconventional T cells in a healthy adult Caucasian population: a combinational study of invariant natural killer T cells, γδ T cells, and mucosa-associated invariant T cells

Abstract: Unconventional T cells show distinct and unique features during antigen recognition as well as other immune responses. Their decrease in frequency is associated with various autoimmune disorders, allergy, inflammation, and cancer. The landscape frequency of the unconventional T cells altogether (iNKT, γδ T, and MAIT) is largely unestablished leading to various challenges affecting diagnosis and research in this field. In this study, we have established the age group–wise frequency of iNKT, γδ T, and MAIT cells… Show more

“…Even without stim, the accumulation of gd T cells were found to be higher in adult compared with newborn (~2.5 fold) and elder (~3 fold, Figure 1B). Age associated alterations in γδ T cell compartment are well studied [71][72][73] . γδ T cells showed an association during viral infections like Cytomegalovirus 74,75 , influenza 76 or COVID-19 [77][78][79] .…”

“…Aging leads to an involution of the function of the immune system which is essentially linked to a lower replicative response to antigenic stimulation and to a decrease in the function of natural killer lymphocyte cells [122][123][124]. Furthermore, with age, we witness the physiological involution of the thymus with a consequent numerical and functional reduction in T-lymphocytes [125,126].…”

“…However, they share some common upregulated and downregulated genes that are different from those in conventional T cells and might provide a clue concerning the relationship between these cells [ 7 ]. MAIT cells recognize the microbial riboflavin-derivative antigens that are presented by the nonpolymorphic MHC class I-like protein MR1 [ 8 ] and are abundant in the liver, representing up to 45% of liver T cells [ 9 ] and up to 10% of T cells in peripheral blood (PB) [ 10 ]; no significant difference was found between sex (male and female) [ 11 ]. The roles of Vα7.2 + /CD161 − T cells are not clearly understood.…”
