Aerosol Delivery of Paclitaxel-Containing Self-Assembled Nanocochleates for Treating Pulmonary Metastasis: An Approach Supporting Pulmonary Mechanics

Abstract: Paclitaxel (PTX) is a potent anticancer agent, which is clinically administered by infusion for treating pulmonary metastasis of different cancers. Systemic injection of PTX is promising in treating pulmonary metastasis of various cancers but simultaneously leads to many severe complications in the body. In this study, we have demonstrated a noninvasive approach for delivering PTX to deep pulmonary tissues via an inhalable phospholipid-based nanocochleate platform and showed its potential in treating pulmonary… Show more

“…All glass twin stage impinger (TSI; British Pharmacopoeia Apparatus A, Copley Scientific, Nottingham, UK) paired with a Copley TPK 2000 critical flow controller linked to a Copley HCP5 vacuum pump [19] was used to check the post nebulization droplet size distribution. The upper (stage I) and lower (stage II) chambers of TSI were filled with methanol, 7 and 30 mL, respectively [20]. The liposomal naringin (1 mg/mL, 5 mL) was aerosolized using an Aeroneb ® Lab micropump nebulizer fitted at the entrance of TSI [21].…”

“…The alveolar epithelium is surrounded by a pulmonary surfactant, which is closely related to the maintenance of normal lung function. [ 56 ] Pulmonary surfactant is a protein‐lipid composed of phospholipids, cholesterol, and proteins, which is similar to the composition of liposomes. [ 57 ] Therefore, the advantages of pulmonary liposome administration are safety, strong compliance, and significant efficacy.…”
