Advances in the Regulation of Macrophage Polarization by Mesenchymal Stem Cells and Implications for ALI/ARDS Treatment

Abstract: Acute lung injury/acute respiratory distress syndrome (ALI/ARDS) is a common condition with high mortality. ALI/ARDS is caused by multiple etiologies, and the main clinical manifestations are progressive dyspnea and intractable hypoxemia. Currently, supportive therapy is the main ALI/ARDS treatment, and there remains a lack of targeted and effective therapeutic strategies. Macrophages are important components of innate immunity. M1 macrophages are pro-inflammatory, while M2 macrophages are anti-inflammatory an… Show more

“…In this study, our prepared EGCG-based hydrogel alleviated ROS accumulation in osteoporotic microenvironment, thus reducing the number of pro-inflammatory M1 macrophages and increasing the number of M2 regenerative macrophages. It is worth mentioning that the effect of Ti-BMSCs@HG group on inducing macrophages to polarize from M1 to M2 is more obvious than that of Ti-HG group, which may be attributed to the immune regulation of loaded rBMSCs in a relieved osteoporosis microenvironment ( Liu C. et al, 2022 ; Shao et al, 2022 ).…”

“…Recently, LPS-induced in vitro and in vivo models have been widely used to study ALI pathogenesis [ 92 ]. M2 macrophages attenuate lung inflammation and inhibit ALI, whereas M1 macrophages promote lung inflammation and contribute to lung tissue injury [ 93 ]. LPS-treated macrophage-derived exosomes impede the cell viability of AECs [ 94 ].…”

“…Late endosomes differentiate into MVBs, which contain intracellular vesicles. Early endosomes develop when the cell membrane is invaginated, and they connect with vesicles created by budding of the Golgi apparatus to form late endosomes 134 . Exosomes can carry proteins, lipids, and miRNAs, which, among other things, help to regulate cellular activity and facilitate intercellular communication 134 .…”

“…Tetraspanins (CD9, CD81, CD63), HSP60, HSP70, TSG101, and Alix are protein markers found on the surface of exosomes 134 . MSC-EVs isolated from the secretome can reproduce the effects of MSCs and regulate macrophage polarization 132, 135 .…”
