Advances in the Development of Antiviral Compounds for Rotavirus Infections

Abstract: Group A rotaviruses (RVAs) are the major cause of severe acute gastroenteritis (AGE) in children under 5 years of age, annually resulting in nearly 130,000 deaths worldwide. Social conditions in developing countries that contribute to decreased oral rehydration and vaccine efficacy and the lack of approved antiviral drugs position RVA as a global health concern. In this minireview, we present an update in the field of antiviral compounds, mainly in relation to the latest findings in RVA virion structure and th… Show more

“…There are more than 685 million cases of acute viral gastroenteritis reported annually [ 74 ]. Viral gastroenteritis (GE), also called “stomach flu”, results in up to 200,000 deaths of children each year, which is one of the highest pediatric mortality rates worldwide [ 75 , 76 ]. Enteric adenoviruses (eAdVs), rotaviruses (RV), astroviruses (HAtVs), and sapoviruses (SaV) are a leading cause of acute GE, along with noroviruses (NoV) [ 74 , 77 , 78 ].…”

“…The GE therapy research is limited, as there are some difficulties with growing certain viruses and/or specific strains [ 85 ]. There are not so many new antivirals and treatments found, as more attention has been on repurposing drugs [ 86 ], such as nitazoxanide [ 87 , 88 , 89 , 90 , 91 ], rupintrivir, and citrate among others [ 76 , 85 , 92 , 93 , 94 ]. However, none of the potential treatments has been introduced as accepted therapy [ 76 ].…”

“…Further antiviral compounds targeting other RV components—e.g., gemcitabine [ 102 ] and 6-thioguanine [ 103 ]—have been reviewed elsewhere [ 97 , 104 ]. Due to limited in vitro results, more research is necessary to characterize their antiviral action.…”

“…A positive impact of RV vaccination has been demonstrated, but the incidence and mortality rate of RV-induced AGE in high- and low-income countries remain to be different from each other. This scenario strongly suggests the necessity of anti-RV strategies to limit the number of deaths deriving from dehydration 5 .…”
