Advances in analytical methodology for bioinorganic speciation analysis: metallomics, metalloproteomics and heteroatom-tagged proteomics and metabolomics

Abstract: The recent developments in analytical techniques capable of providing information on the identity and quantity of heteroatom-containing biomolecules are critically discussed. Particular attention is paid to the emerging areas of bioinorganic analysis including: (i) a comprehensive analysis of the entirety of metal and metalloid species within a cell or tissue type (metallomics), (ii) the study of the part of the metallome involving the protein ligands (metalloproteomics), and (iii) the use of a heteroelement, … Show more

“…6 The coupling of chromatography by using different separation mechanisms with inductively coupled plasma mass spectrometry (ICP-MS) detection has been the most widely used analytical tool for element speciation. [5][6][7][8] In the latest years, fruits and nuts from Amazon region have arisen interest among the scientific community due to the high concentrations of some essential elements in their compositions. [9][10][11][12][13][14][15][16][17] Special attention are devoted to Brazil-nut (Bertholletia excelsa), known as "castanha-doPará".…”

“…The protein standard was prepared dissolving 4.0 mg of ovoalbumin in 2.0 mL of deionized water, submitted to Vortex stirring for 2 min and diluted 10 times with deionized water. The spectrophotometer calibration was performed using analytical reference solutions of 4,6,8,10,12,16 and 20 μg of ovoalbumin in 1.0 mL of Bradford reagent.Water, NaCl, and NaOH extracts of the Brazil-nut samples were diluted 100 times and ethanol extract was diluted 10 times. A volume of 50 μL of these solutions was used for protein determination.…”

“…Mineral compounds are formed and accumulated in the tissues as calcium oxalate, selenium, and nitrates; organic compounds are present in the form of: alkaloids, glycosides, toxalbumins, as well as extremely complex molecules that have not yet been identified (Board et al 2000, Nájera 1993, Szpunar 2005.…”

“…These changes took place in association with new habits of life, especially in highly industrialized western societies, where natural drugs are considPublished: April 07, 2012 plants under certain conditions, and can be of either inorganic or organic nature. “…This is another challenge being increasingly addressed in recent years. [54][55][56][57] In this sequence, two possibilities are described to perform online spike for ID quantification of species by ICP-MS. The basis for quantification of species by ID in flow systems involving different separation techniques coupled to ICP-MS is presented.…”
