Adult human pancreatic acinar cells dedifferentiate into an embryonic progenitor-like state in 3D suspension culture

Abstract: Human pancreatic exocrine cells were cultured in 3D suspension and formed pancreatospheres composed of acinar-derived and duct-like cells. We investigated, up to 6 days, the fate of human pancreatic acinar cells using fluorescein-conjugated Ulex Europaeus Agglutinin 1 lectin, a previously published acinar-specific non-genetic lineage tracing strategy. At day 4, fluorescence-activated cell sort for the intracellularly incorporated FITC-conjugated UEA1 lectin and the duct-specific CA19.9 surface marker, distingu… Show more

“…Another path involves acinar re-programing and trans-differentiation leading to acini-duct metaplasias (ADM) [ 34 , 35 , 36 ]. Both PanIN and ADM processes may be in parallel or in tandem and there might be differences between rodents and humans [ 37 , 38 ].…”

“…The analysis reveals that 444 genes are differentially expressed (p < 0.001), among which we also find biomarkers for the two cell-types (Figure 4(b)-bottom, Figure S1, Supplementary Excel File). Specifically, KRT19, a positive biomarker gene 36 for Ductal cell-type, is significantly underexpressed on the misclassified group and CTRC, a positive biomarker for Acinar cells 36 , is differentially expressed but with a very low expression pattern. These findings suggest that the Ductal cells classified as Acinar are indeed not Ductal, and that further analysis is needed to confirm their true identity.…”

“…These organoids showed the characteristics of ductal cells and a strong proliferative capacity. In addition, one group reported that acinar cells could be dedifferentiated into progenitor-like cells by 3D suspension culture ( Baldan et al, 2019 ). However, whether these progenitor-like cells can be differentiated back into acinar cells remains unclear.…”
