Adrenomedullin Peptide: Gene Expression of Adrenomedullin, its Receptors and Receptor Activity Modifying Proteins, and Receptor Binding in Rat Testis—Actions on Testosterone Secretion1

Abstract: Adrenomedullin (ADM) has been shown to be present in the human and rat male reproductive systems. This study demonstrates the expression of ADM in the rat testis and its effect on the secretion of testosterone. Whole testicular extracts had 5.43 +/- 0.42 fmol of immunoreactive ADM per milligram of protein and 84 +/- 8 fg of ADM mRNA per picogram of Actb (beta-actin) mRNA. Immunocytochemical studies showed positive ADM immunostaining in the Leydig cells and in the Sertoli cells. Gel filtration chromatography of… Show more

“…GPR182 is an adrenomedullin receptor that mediates a cAMP response to the peptide (Kapas et al 1995). Adrenomedullin is reported to be expressed in Sertoli cells and Leydig cells and to inhibit LH/hCG stimulation of testosterone production by the testis (Li et al 2006, Chan et al 2008 although the effect does not appear to be marked. CD36 is a cellular membrane glucoprotein and a member of the class B scavenger receptor family.…”

“…RT-PCR of Adm, Calcrl, Ramp, and Crcp Total RNA of ovary or oviduct was prepared using TRIZOL reagent [18,21] and subjected to real-time RT-PCR. RNA samples (5 lg) were reverse transcribed into cDNA with the SuperScript II reverse transcriptase according to the manufacturer's instructions (Life Technologies, Carlsbad, CA).…”

“…The sample was chromatographed on a Biogel P30 (Bio-Rad Laboratories) column (0.9 3 60 cm), which was eluted with 1 N acetic acid at a flow rate of 1 ml per 10 min for 400 min [18,21]. These 1-ml fractions were assayed for ADM immunoreactivities.…”

“…In the male, ADM is produced in the testis and the male accessory sex glands including the prostate, seminal vesicle, and epididymis. It inhibits steroidogenesis (2,3), stimulates inhibin secretion (4), and increases fluid secretion in the epididymis (5). ADM levels are high in the seminal plasma and ADM can increase sperm motility (6).…”
