Adipose Tissue as a Dedicated Reservoir of Functional Mast Cell Progenitors

Abstract: White adipose tissue (WAT) is a heterogeneous tissue, found in various locations throughout the body, containing mature adipocytes and the stroma‐vascular fraction (SVF). The SVF includes a large proportion of immune hematopoietic cells, among which, mast cells that contribute to diet‐induced obesity. In this study, we asked whether mast cells present in mice adipose tissue could derive from hematopoietic stem/progenitor cells (HSPC) identified in the tissue. We therefore performed both in vitro and in vivo ex… Show more

“…In fact, 1 report demonstrated that murine adipose-derived hematopoietic stem/ progenitor cells contain precursors committed to the mast cell lineage [17]. Another report demonstrated the existence of hemangioblasts in human AT that exhibit the capacity for hematopoietic and endothelial differentiation in vitro [18].…”

“…First, a thorough digestion and disperse is required to obtain purified adipocytes. Fat tissue has complex cellular composition, including adipocytes, ASCs, preadipocyte, fibroblasts, blood cells, endothelial cells, pericytes, smooth muscle cells and so on [3,9,25,[27][28][29][30][31][32]. The purity of derived DFAT cells might be compromised by the contamination of those cells above when they're attached to the floating adipocytes after incomplete enzymatic treatment.…”

“…Los precursores hematopoyéticos del tejido adiposo pueden dar origen a mastocitos, 69 mismos que, además de participar como células fagocí-ticas, tienen un papel esencial en la inflamación. Estas células progenitoras se han propuesto como las posibles causantes de las diferencias entre la hiperreactividad bronquial en modelos animales obesos y no obesos.…”
